摘  要:背景与目的:RSK4是非生长因子依赖的丝/苏氨酸蛋白激酶,参与多种信号通路。本研究构建了人RSK4基因的真核表达系统,研究其对胶质瘤的调控作用,为阐明RSK4功能奠定基础。方法:采用RT-PCR法从人正常乳腺上皮细胞MCF-10A细胞中扩增获得RSK4基因的编码区,定向克隆到真核表达载体pcDNA3.1(-)上,构建真核表达载体pcDNA3.1(-)/RSK4。脂质体法介导pcDNA3.1(-)/RSK4质粒转染至U87细胞中,RT-PCR和Western blot检测表达情况。采用MTT法检测RSK4基因对U87细胞生长的影响。结果:转染pcDNA3.1(-)/RSK4组U87细胞增殖明显被抑制(P<0.05)。结论:RSK4可以抑制胶质瘤细胞的增殖。BACKGROUND OBJECTIVE: RSK4,a class of growth factor-independent threonine/Serinine kinases,plays an important role in siginal transduction.In this study,we constructed an eukaryotic expression system of RSK4 and investigated the role of RSK4 in the growth regulation of U87 cells.METHODS: Human RSK4 eukaryotic expression vector pcDNA3.1(-)/RSK4 was constructed and transfected into human glioma U87 cells.MTT analysis was performed to determine the effect of RSK4 on the proliferation of U87cells.RESULTS: A significant decrease in the proliferation of U87 cells after transfection with pcDNA3.1(-)/RSK4 was observed compared to that in the control groups.CONCLUSON: RSK4 may act as a inhibitor in the proliferation of U87 cells.
