摘  要:目前临床中对宫颈脱落细胞的检查多局限于大体细胞形态学的观察。本文应用原子力显微镜及环境扫描电子显微镜对5例临床宫颈炎患者的宫颈脱落细胞进行了微区力学性质表征以及细胞表面微观形态的成像。结果显示,患者正常形态宫颈上皮脱落细胞在针尖压入深度为700 nm时杨氏模量近似正态分布,峰值在20~30kPa。且细胞表面微嵴明显,微绒毛分布规则。研究表明,AFM可直接应用于临床样品的微区力学性质测定,而环境扫描电镜可以最大限度地展示样品表面的原有形貌特点,为今后临床样品的微尺度物理信息的采集提供了实用的新方法。Nowadays,the cervical exfoliative cytology mainly focuses on general cell morphology in clinic.In this paper,the exfoliated cervical cells from a patient diagnosed as cervicitis were collected.The microscaled mechanical properties of the exfoliated cells were evaluated by atomic force microscopy(AFM),and the topography was imaged by environmental scanning electron microscope(ESEM) in the low vacuum mode.The results show that the young's modulus of the cervical exfoliated cells with normal morphology displayed a normal distribution,and the peak was 500 kPa when the indentation depth was 500nm.The distinct microridges and regularity microvilli were imaged clearly at the cellular surface.The results suggested that the microscaled mechanical properties of clinical samples could be investigated conveniently by AFM,and the original topography should be displayed by ESEM.The AFM and ESEM may be a new method for the evaluation of the microscaled mechanical properties of clinical samples.
