摘　　要：目的探讨博落回提取物(Macleaya Cordataextract,MCE)对大鼠急性酒精性肝损伤的保护作用及其作用机制。方法采用白酒灌胃法建立大鼠急性酒精性肝损伤模型。将60只SD大鼠随机分为模型组,正常组,阳性药物组,MCE高、中、低剂量组,每组10只。检测各组大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)和肝匀浆超氧化物岐化酶(SOD)活性及丙二醛(MDA)含量,观察肝脏病理形态学改变,并测定肝组织中肿瘤坏死因子(TNF-α)、白介素(IL-1β)的表达水平。结果与模型组相比较,经MCE预防性治疗后,大鼠血清中ALT、AST、LDH活性显著降低(P<0.01),肝组织中SOD活力明显增强、MDA含量显著下降、TNF-α与IL-1β表达水平显著降低(P<0.01),形态学观察显示MCE能明显改善肝组织病理形态。结论 MCE能改善肝功能,对大鼠急性酒精性肝损伤具有保护作用,其机制可能与其减轻肝脏炎症、抗脂质过氧化作用有关。Objective To investigate the protective effect and mechanism of Macleaya Cordata extract（MCE） on alcohol-induced acute hepatic injury in rats.Methods Acute hepatic injury model was made by alcohol administration by gavage.Sixty SD rats were randomly divided into 6 groups：a normal control group,a model group,a positive control group,a high-dose MCE group,a middle-dose MCE group,and a low-dose MCE group.The serum ALT,AST,and LDH levels were measured.MDA and SOD in the liver homogenates were analyzed.Histopathological change was observed with hematoxylin and eosin（HE） staining.The expressions of TNF-α and IL-1β in the liver tissues were tested by immunohistochemistry.Results Compared with the model group,after preventive treatment with MCE,the levels of ALT,AST,and LDH in the serum were obviously decreased（P〈0.01）,the activity of SOD in the liver was markedly improved,while the content of MDA and the expressions of TNF-α and IL-1β in the liver were obviously decreased（P〈0.01）.Observation of pathological slices showed MCE improved hepatic pathological changes distinctively.Conclusion MCE can protect the alcohol-induced acute hepatic injury by inhibiting hepatic lipid peroxidation and alleviating the inflammation.
