摘  要:目的:探讨甲基乙二醛(methylglyoxal,MGO)对卵巢癌HO8910细胞生长和增殖的影响。方法:培养卵巢癌HO8910细胞,不同浓度的甲基乙二醛分别处理HO8910细胞不同时间后,用四甲基偶氮唑蓝(MTT)法检测MGO对其生长和增殖的影响,Hoechst33258染色细胞后,荧光显微镜下观察细胞形态学改变;分别以不同的MGO处理HO8910细胞24h,用Anexxin V/PI双染试剂盒染色后经流式细胞仪检测细胞凋亡率。结果:MTT检测发现MGO可抑制细胞生长及增殖,在一定时间和浓度范围内,其抑制率具有浓度依赖性(P<0.01)及时间依赖性(P<0.05);当处理时间大于24h时,其抑制率在浓度为1.5mmol/L时最高,浓度超过1.5mmol/L后抑制率减弱;经MGO处理过的细胞在Hoechst染色后荧光显微镜下呈现典型的凋亡核固缩表现;流式细胞仪检测细胞凋亡率随浓度增高而升高(P<0.001)。结论:甲基乙二醛可显著抑制HO8910细胞增殖并诱导其凋亡,该制剂可能成为卵巢癌新的治疗手段。Objective:To elucidate the effect of methylglyoxal(MGO) on growth and apoptosis of human ovarian cancer cell HO8910.Methods: The human ovarian cancer cells HO8910 were cultured in vitro.HO8910 cells were treated with MGO in different concentrations and times.The effect of MGO on cell growth was evaluated by MTT assay;Hoechst33258 staining assay was used to observe MGO-induced morphological changes;the apoptosis was detected by Flow Cytometer using FITC Annexin-Ⅴ and propidium(PI) staining.Results: As compared with control group,the growth of the cells cultured with MGO was inhibited well marked by MTT assay.The growth inhibition induced by MGO was in a dose-dependent manner(P0.01) when less than 24 hours;however,when the time over 24 hours,the largest inhibition effect was seen at 1.5mmol/L.If the concentration was more than 1.5mmol/L,the inhibition rate decreased.And the growth inhibition was also in a time-dependent manner(P0.05).Apoptosis occurred when HO8910 cells were incubated with MGO,evidenced by morphological changes and increased proportion of Hoechst positive cells.Moreover,apoptotic cells cultured with MGO was of dose dependent that measured by FCM(P0.001).Conclusion: MGO is capable of inhibiting the growth,and it could induce apoptosis in human ovarian cancer cell HO8910.Therefore,MGO could be a potent apoptosis inducer,which may have a potential for ovarian cancer treatment.
