摘 要:目的:探讨热休克蛋白70(HSP70)-1+190(G/C)位点基因多态性对肺癌组织中HSP70-1mRNA及HSP70蛋白表达的影响。方法:应用PCR-RFLP技术检测46例肺癌及相应癌旁正常组织中HSP70-1+190(G/C)位点多态性,RT-PCR法检测HSP70-1mRNA水平,Western blot测定HSP70蛋白。结果:肺癌组织中HSP70-1mR-NA与HSP70蛋白的表达明显高于癌旁正常组织(t=19.968和17.794,P<0.001),并且其表达水平与肺癌的分化程度有关(t=6.813,9.386,P<0.001);肺癌组织中HSP70-1mRNA与蛋白表达水平呈正相关(r=0.817,P<0.001)。46例中,CC型11例(23.9%),GG型17例(37.0%),GC型18例(39.1%),不同基因型携带者HSP70-1mRNA和HSP70蛋白表达水平差异均有统计学意义(F=102.607和62.617,P<0.001),CC基因型携带者表达水平高于GG和GC基因型携带者(P<0.001)。结论:HSP70-1+190(G/C)CC基因型可能是肺癌的一个高危险因素,并能影响其mRNA和蛋白的表达。Aim:To investigate the effect of heat shock protein(HSP)70-1 gene polymorphism on the expressions of HSP70-1 mRNA and HSP70 protein.Methods:PCR-RFLP was applied to detect the single nucleotide polymorphisms(SNPs)of HSP70-1+190(G/C)in 46 patients with lung cancer,RT-PCR was applied to measure the expression of HSP70-1 mRNA,and Western blot was applied to detect the level of HSP70.Results:The expression levels of HSP70-1 mRNA and protein in lung cancer tissue were obviously higher than those in the adjacent normal tissues(t=19.968,17.794,P0.001),and they were related with the differentiation of lung cancer(t=6.813,9.386,P0.001).A highly positive correlation existed between levels of HSP70-1 mRNA and HSP70 protein in lung concer tissue(r=0.817,P0.001).Among the 46 cases,CC,GG and GC genotypes accounted for 23.9%,37.0%,and 39.1%,respectively.There were significant differences in the expression levels of HSP70-1 mRNA and HSP70 protein among the three genotypes(F=102.607,62.617,P0.001).Compared with the other two genotypes,both the levels of HSP70-1 mRNA and HSP70 protein were apparently higher in the individuals with CC genotype(P0.001).Conclusion:CC genotype of HSP70-1+190(G/C)may be a high-risk factor for lung cancer and affect genetic transcription and protein expression.
