摘 要:目的探讨叉头转录因子(FOXO3a)、β-连环素(β-Catenin)、增殖细胞核抗原(PCNA)在乳腺癌发生、发展中的作用。方法选择我院手术切除的乳腺癌组织标本68份(肿瘤组)和乳腺纤维腺瘤组织标本20份(腺瘤组),采用免疫组化Envision两步法检测两组FOXO3a、β-Catenin、PCNA蛋白表达;采用Spearman等级相关分析法分析三指标间及与乳腺癌临床病理参数的相关性。结果肿瘤组、腺瘤组FOXO3a阳性表达率分别为60%9、0%,β-Catenin异常表达率分别为65%、25%,PCNA阳性表达率分别为69%3、5%,两两比较P均<0.05;FOXO3a阳性表达与乳腺癌临床分期、雌激素受体、腋窝淋巴结转移、预后相关,β-Catenin异常表达与乳腺癌组织学分级、腋窝淋巴结转移、临床分期相关,PCNA阳性表达与乳腺癌腋窝淋巴结转移、临床分期相关;乳腺癌组织中FOXO3a阳性表达与β-Catenin异常表达、PCNA阳性表达均呈负相关,β-Catenin异常表达与PCNA阳性表达呈正相关。结论 FOXO3a低表达、β-Catenin异常表达、PCNA高表达与乳腺癌的发生、发展相关。Objectives To investigate the effects of forkbead transcription factor (FOXO3a), β-Catenin and proliferating cell nuclear antigen (PCNA) in the dvelopment and progression of breast cancer. Methods Expression of FOXO3a, β-Catenin and PCNA in 68 patients with breast cancer (tumor group)and 20 patients with breast fibroadenoma ( adenoma group) were examined by immunohistochemical Envision method. Correlation was then analyzed between the expression of three indexes and major clinicopathologic variables. Rttlts The positive expression rates of FOXO3a in the tumor group and adenoma group were 60% and 90%, the positive expression rates of β-Catenin were 65% and 25%, respectively. And the positive expression rates of PCNA were 69% and 35%, repectively. The comparison between the two groups showed that P 〈0.05. The positive expression of FOXO3a in breast cancer was correlated with the clinical staging, ER, node metastasis and prognosis. The abnormal expression of β-Catenin was correlated with the histologic grade, node metastasis and clinical staging. The positive expression of PCNA was correlated with the node metastasis and clinical staging. The positive expression of FOXO3a was negatively correlated with the abnormal expression of β-Catenin and the positive expression of PCNA, and the abnormal expression of β-Catenin was positively correlated with the positive expression of PCNA. Conclusion Low expression of FOXO3a and high expression of β-Catenin and PCNA were correlated with occurrence and development of breast cancer.
