摘  要:目的 探讨Ku80基因在人A549肺腺癌细胞中的表达水平.将A549肺腺癌细胞接种裸鼠,以不同强度的射线照射,探求Ku80蛋白和Ku80 mRNA在放射治疗前后水平变化.方法 利用免疫组化、荧光定量PCR方法检测经过不同强度的射线照射前后的A549肺腺癌细胞的Ku80蛋白含量和Ku80 mRNA的表达.结果 接种在裸鼠的A549肺腺癌细胞经过射线照射后同等时间(24、48h),Ku80蛋白和Ku80 mRNA随着照射强度1、4、8Gy的增强而增加(P<0.05);经过4、8Gy射线照射后48h,测得A549肺腺癌细胞中的Ku80蛋白和Ku80 mRNA较同样强度射线照射后24h而增加(P<0.05).结论 人A549肺腺癌细胞的Ku80蛋白和Ku80 mRNA在一定的照射强度范围内随着照射强度的增强而增加,并在一定的时间范围内随着照射后时间的延长而表达增多,推测Ku80基因可能与肺腺癌细胞放射损伤修复有密切关系.Objective To investigate the changes of Ku80 gene expression in human lung adenocarcinoma cell line A549 before and after radiotherapy. Methods Human lung adenocarcinoma A549 cells were inoculated to nude mice and exposed to different doses of irradiation. The expressions of Ku80 protein and mRNA in A549 cells were detected by immunohistochemistry and real-time Q-PCR, respectively. Results The expressions of Ku80 protein and Ku80 mRNA in A549 cells were significantly increased 24 and 48h after exposure to 1Gy, 4Gy and 8Gy irradiation (P〈0.05). After cells were exposed to 4Gy and 8Gy irradiation for 48h, the Ku80 protein expression was higher than that exposed with the same dose for 24h (P〈0.05). Conclusion The expressions of Ku80 protein and Ku80 mRNA in human lung adenocarcinoma A549 cells are increased after exposure to irradiation in a dose-and time-dependent manner, which suggests that Ku80 gene expression may be involved in the repairing of radiation damages.
