摘  要:目的 探讨精神分裂症及临床特征与肿瘤坏死因子α(TNFα)-G308A多态性之间的相关性.方法 采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)的方法检测57例精神分裂症患者和30例健康对照者TNFα基因G308A位点的基因多态性;用酶联免疫吸附法测定血清TNFα水平;阳性与阴性症状量表评定精神分裂症患者的精神症状.结果 精神分裂症组与对照组的TNFα-308位点GG、GA、AA基因型及 G和A等位基因型频率的差异均无统计学意义(均P >0.05);各基因型患者阳性症状、阴性症状及一般精神病学的总分差异均无统计学意义(均P >0.05);精神分裂症组与对照组血清TNFα水平的差异无统计学意义[(123.6±56.2)pg/ml vs (103.2±66.7)pg/ml,P >0.05],组内TNFα-G308A3种基因型的血清TNFα水平差异无统计学意义(P >0.05).结论杭州地区汉族人群TNFα-G308A多态性和血清TNFα的水平与精神分裂症无明显相关性,也可能对精神分裂症患者的阳性、阴性和一般病理学症状无影响.Objective To analyze the association between the polymorphism of tumor necrosis factor alpha (TNFa) gene promoter G308A and schizophrenia. Methods Fifty seven Han patients with schizophrenia and 30 healthy controls were recruited in this study. TNFa genotypes at G308A were detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), and serum TNF a levels were measured by enzyme-linked immunosorbent assay (ELISA). At the same time, patients were evaluated clinically by the Positive and Negative Syndrome Scales (PANSS). Results There were no significant differences in genotype or allelic distribution of TNF α -308A between schizophrenia patients and healthy controls (P 〉 0.05), and also no statistical difference in PANNS scores among all genotypes (P 〉 0.05). There were no differences in serum TNF α levels between patients and healthy controls [(123.6 ± 56.2)pg/ml vs (103.2 ± 66.7)pg/ml, P〉 0.05],and also no differences in distribution of genotypes between two groups (P 〉 0.05). Conclusion The polymorphism of TNF α -G308A and the serum TNF a levels are not related to schizophrenia or its syndrome scores.
