摘　　要：的　运用肽库筛选技术寻找胃癌单克隆抗体ＭＧｂ２ 所识别的胃癌相关抗原的短肽模拟表位。方法　以胃癌单克隆抗体ＭＧｂ２ 作为筛选配基，对呈现于噬菌体衣壳蛋白ｐⅧ上的两个九肽库进行了亲和富集和免疫筛选；阳性克隆用荧光标记和硝酸纤维素膜斑点印迹证实结合活性；随机抽取部分阳性克隆进行了ＤＮＡ 测序分析。结果　经ＤＮＡ 测序分析推导出相应的氨基酸序列并加以比较分析，得到具有保守性的表位信息。结论　具有相对保守性的ＶＣＸＸＤＧＶ 可能为胃癌单克隆抗体ＭＧｂ２所识别的表位基序。Objective Using a monoclonal antibody against gastric cancer named MGb2 to screen two phage displayed nonapeptide libraries in pⅧ in order to get mimic epitope information. Methods Through affinity enrichment and immunoscreening, positive phages were further identified by fluorescence labeling and dot blot, finally some of the positive clones were sequenced. Results Based on the deduced amino acid sequences after DNA sequencing, some preserved epitope information were obtained. Conclusion VCXXDGV may be a motif which can be recognized by monoclonal antibody MGb2. These results provided useful information for further study of the interaction between antigen and antibody.
