脐血造血干/祖细胞体外培养的实验研究

摘　　要：目的　深入了解脐血的造血功能。方法　采用体外半固体甲基纤维素培养法，检测了脐血中造血干细胞和造血祖细胞的含量。结果　经过１４ ｄ 的体外培养，粒单祖细胞集落为（８６±２６）个，多向造血祖细胞集落为（１３１±７１）个。经过８周的体外培养，起始细胞为（８６±２６）个集落。Objective To study the culture of hematopoietic stem cell/progenitor cell from umbilical blood in vitro.Methods The number of hematopietic stem cell/progenitor cell of umbilical blood were measured by methycellulose colony assay.Results After 2 weeks cultured, CFU GM were 86±26, CFU GEMM were 131±71;After 8 weeks cultured,LTC IC were 86±26.Conclusion The results suggest that umbilical blood transplantation can be establishing hemotopoietic function.
