复方五味子素B逆转人结肠癌细胞THC-8307/OXA多药耐药性机制研究

摘　　要：目的研究复方五味子素B对耐奥沙利铂人结肠癌细胞(TH C-8307/O X A)多药耐药性的逆转作用及其机制。方法采用噻唑蓝(M TT)比色法检测奥沙利铂(O X A)和复方五味子素B(γSC)的细胞毒性;碱性磷酸酶免疫组织化学法和W estern blot检测γSC对TH C-8307/O X A细胞G ST-п蛋白水平的影响。结果γSC(6.0、12.5和25.0μg/m L)对人结肠癌细胞(TH C-8307)和耐奥沙利铂人结肠癌细胞(TH C-8307/O XA)无显著毒性作用(P>0.05),O X A对TH C-8307的IC 50为0.06μg/m L,而对TH C-8307/O X A的IC 50为2.32μg/m L,TH C-8307/O X A是TH C-8307对O X A耐药的39倍,γSC(6.0、12.5和25.0μg/m L)能使O X A对TH C-8307/O X A细胞的IC 50从2.32μg/m L依次下降至0.370、0.128和0.057μg/m L,逆转倍数分别为6.2、18.1和40.7倍。碱性磷酸酶免疫组织化学法和W estern blot检测γSC(12.5μg/m L)处理48 h后,TH C-8307/O X A细胞G ST-п蛋白表达明显降低。结论γSC具有逆转耐奥沙利铂人结肠癌细胞的M D R作用,其作用机制与下调G ST-п表达有关。【Objective】 To study the reversal mechanism of γSC on resistance to OXA in human multidrug-resistance carcinoma of colon cell line THC-8307/OX.【Methods】 The cytotoxic effect of OXA and γSC was evaluated by MTT assay,the effect of γSC on THC-8307/OXA GST-п was evaluated by alkaline phosphatase IHC and western blot.【Results】 MTT assay showed that γSC at the concentration of 6.0,12.5,25 μg/ML has no cytotoxicity to human carcinoma cell line of colon（THC-8307） and its OXA-resistant cell line（THC-8307/OXA）.The IC50 value of OXA to THC-8307 cell line was 0.06 μg/mL and that of to THC-8307/OXA cell line was 2.32 μg/mL,which indicated THC-8307/OXA cell line were 39 times more resistant to OXA in comparison with the THC-8307 cell line,γSC at the concentration of 6.0～25 μg/mL can decrease the IC50 value of OXA to THC-8307/OXA cell line to 0.370,0.128,0.057 μg/mL,reverse multiple was 6.2,18.1,40.7,respectively.GST-П of THC-8307/OXA cell line was significantly decreased through alkaline phosphatase IHC and western blot after treatment with γSC（12.5 μg/mL） and OXA（20 μg/mL） for forty-eight hours.【Conclusion】 γSC can reverse MDR in multidrug-resistant human carcinoma THC-8307/OXA cell line of colon.Its mechanism might be associated with down-regulation of expression of GST-п.
