摘 要:采用传统的平板计数法分析根际土壤可培养细菌、真菌和放线菌数量;同时用聚合酶链式反应-变性梯度凝胶电泳(PCR-DGGE)和BIOLOG方法,对转cry1Ah基因抗虫玉米根部土壤微生物群落结构进行了研究。结果表明,在玉米的整个生长过程中,根部可培养的微生物随时间变化有一定的消长,但同一时间内转基因玉米与非转基因玉米二者根部可培养细菌、真菌和放线菌的数量差异不显著。同时变性梯度凝胶电泳的结果显示,在玉米的整个生长期内,根部土壤细菌群落多样性指数为2.40~2.70,群落分布均匀度指数为0.96~0.99;根部土壤真菌群落多样性指数为2.17~2.48,群落分布均匀度指数为0.95~0.99;转基因玉米和常规玉米根部土壤细菌和真菌的群落结构变化差异并不显著。BIOLOG结果显示,微生物群落的孔平均颜色变化率值、Shannon指数、Simpson指数和McIntosh指数与对照相比无显著差异。以上结果说明cry1Ah基因的导入并没有对玉米根际土壤群落结构产生显著影响。The method of conventional culture techniques was employed to analyze the amounts of bacteria,fungi and actinomycetes in the rhizosphere soil planted transgenic maize with cry1Ah gene.Simultaneously,the effect of microorganism community structure in rhizosphere soil was assessed with culture-independent technique polymerase chain reaction-denaturing gradient gel electrophoresis(PCR-DGGE) and BIOLOG assay.During the growth process of the maize,the amount of cultured rhizosphere microorganisms was variation,but no significant difference between transgenic and regular maize under the same period.Moreover,the results of cluster analysis of DGGE profile of bacteria and fungi indicated that there was no remarkable difference in the same phase.For the two investigations,the Shannon′s index of the bacterial community in the rhizosphere soil was 2.40-2.70 and the evenness index was 0.96-0.99;the Shannon's index of the fungal community in the rhizosphere soil was 2.17-2.48 and the evenness index was 0.95-0.99.BIOLOG assay results showed that there was no remarkable difference of the average well color development,Shannon,Simpson and McIntosh of between transgenic and common maize.It implied that the transgenic maize with the cry1Ah gene did not affect significantly on community structure of microorganism in rhizosphere soil.
