大黄素对人胃癌细胞MKN45增殖抑制及诱导凋亡的研究

摘　　要：目的:研究大黄素(Emodin)对人胃癌细胞MKN45增殖及诱导凋亡的作用。方法:用噻唑蓝(MTT)法观察Emodin对MKN45的生长抑制作用;用DNA Ladder凝胶电泳法、荧光染色法、流式细胞仪法分析Emodin的诱导凋亡作用。结果:与对照组相比,Emodin能明显抑制MKN45细胞的生长且与浓度、时间呈依赖性,其IC50(48小时)为(47.5±0.3)μmol/L;Emodin处理胃癌细胞后,DNA凝胶电泳结果显示呈明显的梯状条带;吖啶橙(AO)染色观察示细胞核内可见浓染致密的颗粒荧光、新月型改变、核固缩或片段化及凋亡小体的形成;流式细胞仪分析Emodin能浓度依赖性诱导MKN45细胞凋亡和G2/M期阻滞。结论:Emodin对人胃癌细胞MKN45有明显的生长抑制作用,且该作用与诱导细胞凋亡有关。Objective：To study emodin on proliferation and induced apoptosis of human gastric cancer cell MKN45.Method：The growth inhibition of Emodin on MKN45 was observed by MTT;analysis of Emodin-induced apoptosis by DNA Ladder gel electrophoresis,fluorescence staining and flow cytometry.Result：Compared with control group,Emodin significantly inhibited the growth of MKN45 cells in a dose and time dependent,the IC50（48 hours） （47.5±0.3）μmol/L;after treatment of Emodin on gastric cancer cells,DNA gel electrophoresis results showed a clear laddering;acridine orange（AO） staining showed the nucleus visible fluorescent stain dense granules,crescent change, nuclear condensation or fragmentation and apoptotic body formation,flow cytometry analysis of Emodin concentration-dependent induction of apoptosis in MKN45 cells and G2/ M phase arrest.Conclusion：The growth of human gastric cancer cells MKN45 significantly was inhibited by Emodin,and the effect was related to apoptosis.
