多效蛋白PTN在肺腺癌血清、组织和胸水细胞中的表达及意义

摘　　要：目的通过检测肺腺癌患者术前血清标本及相对应的术后切除肺癌组织标本,和肺腺癌转移胸水标本中的PTN蛋白的表达,探讨PTN蛋白在肺腺癌患者三种不同标本中的表达及意义。方法利用Western-blot、免疫组织化学、免疫细胞化学方法分别检测189例上述三种不同标本中PTN蛋白的表达,其中有29例对应的术前血清,肺腺癌术后组织和术后3-4个月的转移胸水标本,同时分别以50例正常献血者血清、50例来自于189例肺腺癌癌旁组织和50例胸水良性增生细胞作为对照。结果肺腺癌患者血清、组织、胸水中PTN蛋白的表达分别高于对照组PTN蛋白的表达。PTN蛋白在肺腺癌患者胸水细胞中的表达率57.7%(109/189)高于组织中的表达率42.9%(81/189)、组织中的表达率42.9%(81/189)高于肺腺癌患者血清中的表达率18%(53/189),差异均具有统计学意义(P<0.05)。在189例肺腺癌患者的胸水标本中PTN和Vim-entin的表达呈正相关关系(P<0.01,r=0.728)。并且,在29例分别来源于同一例患者的这三种标本中,27例PTN在血清和肺腺癌组织中阴性表达,但在相对应的恶性胸水腺癌细胞中PTN呈阳性表达。同时,在这27例对应的标本中,PTN阳性表达的标本Vimentin亦阳性表达,而E-ca阴性表达。结论 PTN蛋白在肺腺癌患者血清、组织和胸水中高表达,且PTN和Vimentin的表达一致说明PTN在肺腺癌的EMT转化过程中可能起到重要的作用。Objective To detect the expression of PTN protein in the serum of lung adenocarcinoma patients before surgery,their corresponding lung adenocarcinoma tissue after surgery,and lung adenocarcinoma cells from pleural fluid and to discuss the relationship and significance.Methods The expression of PTN protein in three kind of specimens（totally 189） was detected by Western-blot,immunohistochemistry and immuncytochemistry.There were 29 patients from whom serum before surgery,carcinoma tissue after surgery,and carcinoma cells from pleural fluid were all taken.Meanwhile,normal blood serum,normal tissues and bengin pleural fluid from 50 cases each served as controls.Results The positive rates of PTN protein in the serum corresponding carcinoma tissue,and carcinoma cells from pleural fluid were respectively higher than that of their controls.The positive rate of PTN in pleural fluid（57.7%,109/189）was higher than that in tissue（42.9% 81/189）,and the rate of in tissue was higher than that in blood serum（18% 53/189）,with statistical significance（P0.05）.The expression of PTN and Vimentin had a positive correlation in carcinoma cells from pleural fluid of 189 patinents（P0.01,r=0.728）.Moreover,in the 3 different specimens of the 29 patients,27 cases showed PTN negative expression in serum and carcinoma tissues but positive expression in corresponding carcinoma cells from pleural fluid.At the same time,the expression of PTN was consistent with that of Vimentin but contrary to that of E-ca.Conclusion PTN is highly expressed in serum、 tissue and pleural fluid of lung adenocarcinoma patients.The consistent expression of PTN and Vimentin suggests that the expression of PTN in lung adenocarcinoma has a significance relationship with,epithelial-mesenchymal transition of the tumor.
