摘 要:目的研究塞来昔布对人结肠癌细胞系HT-29增殖的抑制作用及其对HT-29细胞微小核糖核酸(mi-RNA)表达谱的影响。方法采用CCK-8比色还原法观察塞来昔布对HT-29细胞增殖的抑制作用;miRNA芯片技术检测塞来昔布作用前后HT-29细胞miRNA的表达变化,实时定量RT-PCR验证其结果。结果塞来昔布对HT-29细胞增殖有显著的抑制作用,且呈剂量时间效应关系,其半数抑制浓度为(237.73±1.65)μmol/L;塞来昔布作用于HT-29细胞48 h后,获得28个与塞来昔布干预相关的miRNAs(P<0.01),其中8个下调,20个上调。结论塞来昔布对miRNA表达谱的影响可能是其抑制HT-29细胞株生长的作用途径之一。Objective To study the anti-tumor effect of celecoxib on HT-29 colon cancer cell in vitro and its effect on the expression profiles of microRNA(miRNAs).Methods Human colon cancer cells HT-29 was treated by celecoxib in various concentrations alone.The inhibitory effect of cell growth was assayed by CCK-8 method.The expression profiles of miRNA was determined using miRNA microarray.The expression of selected miRNA was then validated by real-time RT-PCR.Results The inhibitory effect of celecoxib on the proliferation of HT-29 cell was found in does-and-time dependent manner.The IC50 value was(237.73±1.65)μmol/L,after 48 h,twenty-eight miRNA,20 up-regulation and 8 down-regulation expression with statistical significance were identified in colon cancer cells following exposure to celecoxib compared to matched control cells.Conclusion Celecoxib could inhibit the HT-29 cell growth in vitro partly attributting to its effect on the miRNA expression.
