摘  要:目的 ;建立GL261胶质瘤细胞C57BL/6小鼠、C6胶质瘤细胞SD大鼠及BALB/c小鼠皮下动物模型,比较其肿瘤生长特点。方法借助动物立体定向仪,将体外培养小鼠GL261、大鼠C6胶质瘤细胞分别接种于C57BL/6小鼠及SD大鼠右侧尾状核区,C6胶质瘤细胞接种于BALB/c小鼠左前肢皮下。接种后观察不同种实验鼠的生存状态及肿瘤的生长特性,颅内模型于接种后7d、14d、21d、28d进行MRI检查,皮下模型测量体积,并绘制生长曲线。解剖标本,做组织病理学和胶质纤维酸性蛋(GFAP)免疫组化检查。结果 GL261胶质瘤细胞C57BL/6小鼠模型较之后两种模型在组织病理学上接近人脑胶质瘤,而且颅内生长稳定,成瘤率高,未见颅外转移病灶,实验周期短,重复性好。结论 GL261胶质瘤细胞C57BL/6小鼠模型,其肿瘤生长特性及病理特征与人脑胶质瘤相似,可作为临床胶质瘤基础研究的理想模型。Objective To explore tumor growth characteristics of mouse glioma GL261 model and SD,BALB/c C6 glioma models.Methods The glioma cells cultured in vitro were stereotaxically implanted into right caudate nucleus of C57BL/6 mouse and SD rats,C6 cells were implanted into subcutaneous of BALB/c mouse.The living and survival states were Observed.After inoculation,use an MRI protocol to evaluate intracranial tumor growth on 7,14,21,28 day.Measure tumor volume of subcutaneous model,and draw the growth curve.Animals were killed for the study of tumor histopathology.Tumor was confirmed with HE and staining of glial fibrillary acidic protein(GFAP) immunohistochemistry.Results Mouse glioma GL261 model was more similar to histopathological features of human glioma.This kind of model was a more reliable and reproducible one,with 100% yield of intracranial tumor and no extracranial metastasis.Conclusion Mouse glioma GL261 model resembles histopatho-logical features of human glioma,which can be used as gliomas a ideal model for etiology research of glioma.Whereas further exploration is required for the other two models.
