摘 要:目的探讨蛇毒精氨酸酯酶Agkihpin对人鼻咽癌CNE-2细胞系环氧合酶-2(COX-2)表达的影响。方法用不同浓度的Agkihpin处理鼻咽癌CNE-2细胞系72 h后,应用免疫细胞化学、Western Blotting、反转录-聚合酶链反应(RT-PCR)法检测COX-2在CNE-2细胞系中的表达。结果不同浓度Agkihpin处理CNE-2细胞系72h后,COX-2的平均吸光密度比较,差异有统计学意义(P<0.05);其中A1组、B1组和C1组较D1组均明显降低,差异有统计学意义(P<0.05),A1组较B1组、C1组和D1组均明显降低,差异有统计学意义(P<0.05)。WesternBlotting检测经不同浓度Agkihpin处理72 h后COX-2表达差异有统计学意义(P<0.05),随着Agkihpin浓度的降低,COX-2表达水平升高,各组间比较差异均有统计学意义(P<0.05)。不同浓度Agkihpin处理72 h后COX-2 mRNA的相对转录水平比较差异有统计学意义(P<0.05),随着Agkihpin浓度的降低,COX-2 mRNA的相对转录水平升高,各组间比较差异均有统计学意义(P<0.05)。结论在人低分化鼻咽癌CNE-2细胞系中,Agkihpin能抑制COX-2的表达,并且随着浓度的增大更明显;Agkihpin对COX-2的抑制作用,可能成为鼻咽癌化疗的有效药物。Objective To explore the impact of Agkihpin on the expression of cyclooxygenase -2 in nasopharyngeal carcinoma CNE -2 cell line and to describe the mechanisms of Agkihpin inhibition against CNE -2. Methods The cultured cells were treated with different concentrations of Agkihpin for 72 h. The expression levels of COX - 2 in CNE - 2 cell line were assayed by immunocytoehemistry, Western blotting, and RT - PCR. Results The average COX - 2 absorbance among different Agkihpin concentrations showed significant differences after 72 hours of treatment ( P 〈 O. 05 ) ; more specificly, the measurement was low- er in group A, B, C than in group D ( P 〈 0. 05 ) and lower in group A than in group B, C, D (P 〈 0. 05 ) . Western Blotting showed significant differences among different Agkihpin concentrations after 72 hours of treatment ( P 〈 0. 05 ) ; COX - 2 expres- sion increased as Agkihpin concentration decreased ( P 〈 0.05 ) . Expression levels of COX - 2 mRNA showed significant differ- ences among different Agkihpin concentrations after 72 hours of treatment ( P 〈 0.05 ) ; COX - 2 expression increased as Agkihpin concentration decreased ( P 〈 0. 05 ) . Conclusion Agkihpin inhibits the expression of COX - 2 in CNE - 2 cells in a dose - de- pendent manner, which is likely one of the reasons for Agkihpin inhibition of the cellular vitality of nasopharyngeal carcinoma cells; Agkihpin might be applied in the treatment of nasopharyngeal carcinoma.
