摘 要:目的:探讨新型重组高效复合干扰素对卵巢癌HO8019细胞的作用,并与同类型复合干扰素、普通重组Ⅰ型干扰素、化疗药物对比。方法:应用新型重组高效复合干扰素、复合α干扰素、干扰素α-2b和顺铂以0.156ug/m l、0.625ug/m l、2.5ug/m l及10ug/m l浓度作用于体外培养的HO8019细胞72小时后,MTT法检测并计算细胞抑制率并比较。应用新型重组高效复合干扰素、复合α干扰素、干扰素α-2b和顺铂以0.156 ug/m l、0.625 ug/m l、2.5 ug/m l浓度作用于体外培养的HO8019细胞72小时后,以流式细胞仪检测HO8019细胞的凋亡率并进行比较。结果:几种干扰素及顺铂均能导致HO8019细胞生长抑制及促进细胞凋亡。相同浓度条件下,新型重组高效复合干扰素和顺铂对HO8019细胞的抑制及凋亡作用强于复合α干扰素和干扰素α-2b的作用,尤以顺铂的作用最强。结论:重组高效复合干扰素对体外培养的卵巢癌HO8019细胞的生长有抑制作用,并能促进HO8019细胞凋亡;作用虽然不及化疗药物顺铂,但较同类型干扰素、普通重组Ⅰ型干扰素的作用强。Objectives: To investigate the effect of rSIFN-co consensus Interferon,IFNa-2b(Interferon a-2b) and chemotherapeutics on ovarian cancer cell line HO8019 in vitro.Methods: Αfter HO8019 cervicαl cαncer cells were cultured in vitro with rSIFN-co、Infergen、IFNα-2b αnd cisplαtin(DDP)αt the concentrαtion of 0.156ug/ml、0.625 pg/ml、2.5 ug/ml and 10 g/ml respectively for 72 hours,cell proliferαtion was mensurated by MTT assay.Cell inhibition rate was calculated.Then after HO8019 ovarian cancer cells were treated with rSIFN-co、Infergen、IFNa-2b and DDP at the concentration of 0.156 ug/ml、0.625 pg/ ml、2.5 pg/ml for 72 hours,cell apoptosis wαs determined by flow cytometry.Results: rSIFN-co、Infergen、IFNα-2b and DDP could induce inhibition and apoptosis of HO8019 cells.The inhibition and apoptosis of HO8019 cells which had been induced by rSIFN-co and DDP were better than which had been induced by Infergen and lFNα-2b at the same concentration,and DDP was more better than rSIFN-co.Coclusions: rSIFN-co has antineoplastic efect by inhibiting proliferation and inducing apoptosis in HO8019 cervica cancer cell in vitro.The effect is inferior to DDP,but better than the same type interferon and general type I interferon.
