摘  要:目的:建立阿霉素诱导的大鼠肾纤雏化模型并观察其病理变化。方法:利用单侧肾切除术加2次阿霉素尾静脉注射法诱导大鼠出现肾病综合症的表现,常规组织病理学方法观察肾组织的病理改变。结果:光镜下可见模型组大鼠肾小球节段性硬化和玻璃样变,硬化的肾小球周围所属肾小管萎缩、基底膜增厚,部分肾小管消失;部分肾小球肥大周围肾小管扩张严重;肾小球集中现象明显;肾小管上皮细胞变性、坏死,蛋白管型严重;肾间质胶原纤维增生和大量炎细胞浸润。伴有尿蛋白增加、水肿、低体重,以及肾功能异常等表现。结论:单侧肾切除术加2次阿霉素尾静脉注射法诱导大鼠的肾纤维化病变典型,是研究肾纤维化较为理想的动物模型。Obstractive: To establish an Adriamycin induced rat nephrosis model and observe its pathological changes. Methods: The rat Adriamycin nephrosis mode was established by injecting Adriamycin into tail - vein twice , 8 weeks end to detect blood and urina biochemical parameters and to observe the pathological changes of renal tissues. Blood serum creatinine (SCr) and blood urea nitrogen(Bun) content were determined by colorimetric method. Histological changes of kidney were observed with hemotoxylin and eosin staining. FN,LN were observed by immunohistochemistry method. Results: Pathological examination of kidney in the model group showed thickened glomeruar/tubular basement membrane with segmental sclerosis and hyaline degeneration ; atrophy of the renal tubule around the sclerotic glomeruli and part of them disappeared;hypertrophy of partial glomeruli with surrounding severe dilated tubules;obvious glomemli centering phenomena; severe tubular epithelial cell degeneration, necrosis with protein cast;fibrous tissue proliferation and large amount of inflammatory cell interstitial infiltration. In mod- el rats, the levels of total urinary protein in 24 h, BUN, SCr were significantly higher and body weight lower than those in the control group ( P 〈 0.05 ). Conclusion:The rat Adriamycin nephrosis mode established by injecting Adriamycin into tail -vein twice was an ideal renal fibrosis model.
