摘 要:目的:分析福州地区甲型H1N1流感病毒株的全基因序列和遗传特征。方法:采用MDCK细胞培养法和Real-time RT-PCR法对A/Fuzhou/721/2009(H1N1)进行病毒分离、鉴定。测定该株的全基因组序列,并进行基因进化树分析。结果:分离株的8个基因片段核苷酸和推导氨基酸序列与其他地区新甲型H1N1参考株的同源性都在99%以上;与参考株A/California/04/2009相比,HA蛋白有4个氨基酸发生了突变,分别位于83、1972、03和321位点上,HA1蛋白都有6个潜在糖基化位点;对耐药性位点分析发现,对金刚烷胺类药物耐药,但仍然对神经氨酸酶抑制剂类药物敏感。结论:该毒株与2009年大流行株高度同源;相对于以往古典型猪流感代表株出现了HA蛋白抗原性漂移,但HA蛋白保留了古典猪H1的特点。对福州地区甲型H1N1(2009)流感病毒全基因序列和分子遗传特征的详细研究,为监测病原变异、致病性及流行规律提供科学依据。Objective:Influenza type A H1N1 virus was isolated from the confirmed cases with influenza in Fuzhou and its whole-genome sequencing and genetic characteristics were analyzed.Methods:A/Fuzhou/721/2009(H1N1) were isolated and identified by MDCK cell culture and Real-time RT-PCR.Then complete genome sequence of the strains were determined,and analyzed using phylogenetic tree.Results:Phylogenetic analysis confirmed that each of the 8 genes of virus strain was highly homologous with current new H1N1 virus originated in the influenza pandemic in 2009,with homology above 99% Compared to reference virus A/California/04/2009,the HA protein of the isolate had four amino acid mutation,and the isolate contained six potential glycosylation sites in HA1.Analysis of amino acid sequences indicated that this virus possessed oseltamivir sensitivity and amantadine resistibility.Conclusion:This virus strain was highly homologous with pandemic strains in 2009.In comparison with the representative strains of classic swine flu,HA antigenic draft were found,but retaining the features of classic swine H1.The detailed research on the virus strains in Fuzhou provided scientific basis for the surveillance of pathogen mutation,pathogenicity and epidemic rule.
