摘 要:目的探讨microRNA(miRNA)-148a对前列腺癌细胞系LNCaP神经内分泌分化的影响。方法采用实时定量聚合酶链反应(RT-PCR)验证miRNA-148a在雄激素依赖性与雄激素非依赖性前列腺癌细胞系中的差异表达。采用miRNA-148a及其抑制物(anti-miRNA-148a)改变LNCaP细胞中miRNA-148a的表达量,观察LNCaP细胞神经内分泌分化的差异程度,并检测神经内分泌分化标志物神经元特异性烯醇化酶(NSE)的差异表达。结果 LNCaP-AI细胞中的miRNA-148a相对表达量显著低于LNCaP细胞(P<0.05)。下调miRNA-148a在LNCaP细胞中的表达量,可明显促进LNCaP细胞神经内分泌分化。结论 miRNA-148a可以抑制LNCaP细胞的神经内分泌分化,可能成为前列腺癌生物治疗的新靶点。Objective To detect the effect of microRNA-148a on the neuroendocrine differentiation of human prostate cancer cell line LNCaP. Methods The differential expression of microRNA (miRNA-148a) in androgen-dependent prostate cancer (ADPC) and androgen-independent prostate cancer (AIPC) cell lines was examined by real time (RT)-PCR. miRNA-148a and anti-miRNA-148a were used to affect the expression of miRNA- 148a in LNCaP cells. Neuroendocrine differentiation of LNCaP cells was observed under inverted microscope. The level of neuron-specific enolase (NSE), a marker of neuroendocrine differentiation, was detected by RT-PCR. Results RT-PCR results showed that LNCaP-AI cells had a significantly lower expression of miRNA-148a compared with LNCaP cells (P〈0. 05). We also found that down-expression of miRNA-148a promoted the neuroendocrine differentiation of LNCaP cells. Conclusion miRNA-148a can inhibit neuroendocrine differentiation in LNCaP cells, which might be a promising target for biotherapy of prostate cancer.
