摘 要:目的比较表皮生长因子(EGF)和碱性成纤维细胞生长因子(bFGF)对体外人视网膜色素上皮细胞(hRPE)增殖调控的剂量-效应关系、时间-效应关系,从而筛选出在体外培养hRPE增殖效应较强的生长因子,以提高体外hRPE的培养成效。方法采用经细胞化学染色法鉴定确认的hRPE细胞株,用CCK-8法检测成组配对的EGF和bFGF(0、2、4、8、16、32 ng/ml)在不同作用时间(0、24、48、72、96 h)对体外hRPE增殖的影响。结果在相同浓度EGF或bFGF作用下,hRPE吸光度值(A值)随作用时间延长而增加,与对照组比较的差异有统计学意义(P<0.05)。在相同培养时间条件下,hRPE A值随EGF或bFGF浓度增加而增加,与对照组比较的差异有统计学意义(P<0.05);在相同作用时间下,浓度为4 ng/ml以上时,EGF组的hRPE吸光度值(A值)比bFGF组高且差异有统计学意义(P<0.05)。结论 EGF和bFGF对体外培养hRPE增殖的调控有剂量-效应关系和时间-效应关系。同浓度EGF的促进细胞增殖效应强于bFGF。Objective To compare the dose-response and time-effect relationship of cell proliferation regulation of the epidermal(EGF) and the basic fibroblast growth factor(bFGF)on human retinal pigment epithelium(hRPE) cultured in vitro,and find the stronger growth factor on the proliferation of hRPE cultured in vitro to improve the effectiveness in vitro cultivation of hRPE.Methods CCK-8 was used to detect the effects of pairs of EGF and bFGF with different concentration(0,2,4,8,16,32 ng/ml) on the hRPE proliferation at different times(0,24,48,72,96 h).Results Cultured under the same concentration of EGF or bFGF,hRPE absorbance(A value) increases with time.Compared with the negative control group,the difference was statistically significant(P0.05).Cultured during the same time,hRPE absorbance(A value) increases with the rise of the concentration of EGF or bFGF.Compared with the negative control group,the difference was statistically significant(P0.05).Cultured under the same time,the proliferation effect of EGF group was stronger than its corresponding bFGF group when the concentration was above 4 ng/ml and the difference between them was statistically significant(P0.05).Conclusion Certain dose-response relationship and time-effect relationship exist in the regulation of EGF and bFGF on cell proliferation of hRPE cultured in vitro.The effect of EGF is stronger than effect of bFGF on culturing RPE cell proliferation.
