放化疗对ezrin蛋白在宫颈鳞癌中表达的影响及意义

摘　　要：目的探讨放化疗对宫颈鳞癌组织中ezrin蛋白表达的影响及意义。方法应用免疫组化SP法检测20例正常宫颈上皮(normal cervical epithelium NCE)、36例宫颈上皮内瘤变(cervical intraepithelial neoplasm CIN)、40例宫颈鳞癌(squamous carcinoma of cervix SCC)在放化疗前(SCC-pre)、放化疗后(SCC-post)中ezrin蛋白表达。结果 ezrin蛋白在NCE主要表达于细胞膜,而在CIN和SCC主要表达于细胞质。ezrin蛋白阳性率从NCE到CIN,再到SCC,ezrin蛋白阳性率显著升高(P<0.05)。放化疗后ezrin蛋白在宫颈鳞癌表达的强阳性率显著性降低(P<0.05)。ezrin蛋白在SCC的阳性表达分别与间质浸润深度及脉管浸润有关(P<0.05),而与组织学分级及盆腔淋巴结转移无关(P>0.05)。结论 ezrin蛋白在宫颈鳞癌发生发展及浸润转移中可能发挥重要作用,放化疗可抑制ezrin蛋白表达。Objective To investigate the effect and significance of chemoradiation to the expression of ezrin protein in squamous carcinoma of cervix（SCC）.Methods The expression of ezrin protein were respectively detected by immunohisto-chemistry in 20 cases of normal cervical epithelium（NCE）,36 cases of cervical intraepithelial neoplasm（CIN）,40 cases of squamous carcinoma of cervix for pre-and post-preoperative radiotherapy and neoadjuvant chemotherapy.Results The positive expression of ezrin protein gradually increased from NCE to CIN,and then to SCC group（allP0.5）.In the cases with dee-per stroma involvement and intravasclar infiltration,the expression of ezrin was significantly higher than those in the cases with-out them（allP0.05）.In the NCE,the positive staining reaction of ezrin protein were mainly located in the cell membrane;in the CIN and SCC groups,the positive staining reaction mainly located in the cell endochylema,the staining reaction weakened or vanished in cell membrane.Conclusion The over expression of ezrin protein is correlated with the squamous carcinoma of cervix development,invasion.Radiotherapy and chemotherapy could down-regulate the level of ezrin protein in squamous carci-noma of cervix.
