摘  要:目的研究微囊化人胰腺癌细胞建立裸鼠胰腺癌模型效果,以期建立稳定的更为理想的胰腺癌动物模型。方法分别以人胰腺癌细胞悬液和微囊化人胰腺癌细胞悬液建立皮下移植瘤模型,定期监测皮下肿瘤大小,绘制生长曲线并进行比较。分别以人胰腺癌细胞悬液和微囊化人胰腺癌细胞建立原位移植瘤模型,分别于模型建立后第4周和第8周进行正电子发射计算机断层显像检查及剖腹探查,以评估肿瘤生长及转移情况,标本作电镜及病理检查。结果细胞悬液组和微囊悬液组皮下移植瘤模型在成瘤体积和成瘤率方面无明显差异。微囊化肿瘤细胞法建立原位移植瘤模型较肿瘤细胞悬液法在成瘤质量及出现转移灶等方面有较强的优势。两组所形成的原位移植瘤在病理学方面无明显差异。结论微囊化肿瘤细胞法是一种可靠的有发展前景的建立胰腺癌动物模型的方法。Objective To establish a stable and ideal nude mice model of pancreatic cancer by evaluating the result of encapsulation of pancreatic tumor cells. Methods Two kinds of subcutaneous implantation models were established by injection of single tumor cells and encapsulated pancreatic tumor cells. Inspected the size of subcutaneous implantation tumors established by two methods on a weekly basis, draw the growth curve and compared the result. Evaluated the growth and metastasis of orthotopically injected single tumor cells and encapsulated pancreatic tumor cells by PET-CT scan and necropsy at fourth and eighth week after implantation. The pancreatic tumor specimens attained by two methods was taken for pathological examinations. Results In subcutaneous implantation model, there was no statistical difference observed between encapsulated pancreatic tumor cells and single tumor cells in terms of tumor volume and rate of tumor incidence. The former method has obvious advantage in tumor mass and rate of metastasis compared with the latter one in the orthotopic implantation model. Conclusion Encapsulation of pancreatic tumor cells is a reliable and prospective method of establishing pancreatic tumor animal model.
