摘　　要：目的探讨苯乙酸钠(sodium phenylacetate,NaPA)联合热疗诱导人喉癌细胞株(Hep-2细胞)的增殖抑制率、细胞凋亡率及其对细胞周期进程的影响,旨在为喉癌的综合治疗提供理论依据。方法应用MTT比色法检测不同条件下对Hep-2细胞的增殖抑制率,流式细胞仪检测细胞周期进程的变化,荧光显微镜检测Hep-2细胞的细胞凋亡率。结果单纯热疗组、NaPA组及联合组对Hep-2细胞的抑制率分别为24.8%,27.5%和45.3%,与对照组相比差异显著,P<0.01;流式细胞仪结果显示G1/G0期细胞增多,S期细胞减少,G2/M期细胞相对增多,组间比较差异显著;荧光染色结果:单纯热疗组与NaPA组细胞凋亡率无差异,但联合组效果明显。结论 NaPA联合热疗能显著提高Hep-2细胞的增殖抑制率,抑制Hep-2细胞G1期向S期转化进程,诱导Hep-2细胞凋亡。Objective To study sodium phenylacetate（NaPA） associated with hyperthermia induced laryngeal carcinoma cell line（Hep-2 cells） proliferation inhibition ratio,apoptosis rate and its effect on cell cycle progression,for comprehensive therapy of laryngeal carcinoma provide some theoretical basis.Methods MTT colorimetry detected laryngeal carcinoma cell line（Hep-2 cells） proliferation inhibition ratio in different conditions;Flow cytometry detected the variety cell cycle progression;Fluorescence microscope detected laryngeal carcinoma cell line（Hep-2 cells） apoptosis rate.Results The inhibition ratio of single hyperthermia group,NaPA group and combined group were 24.8%、27.5% and 45.3%.The difference of control group was significant,（P0.01）.The results of flow cytometry show number of cell in G1/G0 phase increasing,M phase decrease,G2/M phase relative increase.Significant difference each groups.The result of Staining：the apoptosis rate of single hyperthermia group and NaPA group was no difference,however,the difference of combined group was significant.Conclusion NaPA associated with hyperthermia could significantly improve laryngeal carcinoma cell line（Hep-2 cells） proliferation inhibition ratio.,and inhibite Hep-2 cells from G1 to S phase in the transformation process,induce Hep-2 cells apoptosis.
