摘  要:目的:研究青钱柳多糖的抗氧化活性。方法:采用水提醇沉方法,Sevage法除蛋白,用D301R型大孔树脂纯化制备多糖;通过Fenton体系,DPPH·两种分析方法测定体外清除自由基能力;用青钱柳多糖对高脂小鼠进行干预后,测定肝脏SOD、MDA、GSH-Px、NEFA的含量,以此评定其体内抗氧化功能。结果:青钱柳多糖对羟基自由基、DPPH自由基清除效果显著,并且清除DPPH的效果优于羟基自由基;体内实验显示:青钱柳多糖能显著提高肝脏SOD和GSH-Px的活性,降低肝脏MDA及NEFA的含量。结论:体内及体外实验都显示青钱柳多糖具有显著的抗氧化活性。Objiective:To study the anti-oxidant activity of polysaccharide from Cyclocarya paliurus(Batal.)Iljinsk.Methods:The polysaccharide from Cyclocarya paliurus(Batal.) Iljinsk was extracted by water extracting-alcohol precipitating process,and the protein was removed by Sevage method.Then polysaccharide was isolated and purified by D301R column chromatography.The radical remove-ability of the polysaccharide was tested by DPPH and Fenton methods in vitro.The Hyperlipidemia mice were intervened with polysaccharide from Cyclocarya paliurus(Batal.) Iljinsk.To assess its antioxidant function in vivo,levels of lipids,activity of superoxide dismutase(SOD),activity of glutathione peroxidase(GSH-Px),levels of malondialdehyde(MDA) and free fatty acids(NEFA) in the liver were quantitatilatively measured.Results:It was demonstrated that the polysaccharide had significant scavenging activities of hydroxyl radical and DPPH radical at different digrees,and the ability of scavenge DPPH radical is higher than that of the hydroxyl radical in vitro.Also,the polysaccharide significantly increased SOD and GSH-Px activities,decreased MDA and NEFA contents in the liver.Conclusion:The results showed that the polysaccharide from Cyclocarya paliurus(Batal.) Iljinsk has significant antioxidant activity in vivo and in vitro.
