摘 要:以牛睾丸为原材料,采用过滤离心相结合的方法提取透明质酸酶。研究了提取剂、静置时间、分离方式、盐析试剂及盐析试剂加入量对透明质酸酶活力的影响。结果表明,较佳提取条件为:以pH 3.5的盐溶液(由0.1 mol/L乙酸+1 mmol/L盐酸+0.1 mol/L NaC l配成)为提取剂,提取液0℃静置8 h。提取液过滤后3 000 r/m in离心分离,加入240 g/L硫酸铵作为盐析试剂,此时酶活力较高,为277.5 U/mL,比活为12.4 U/μg,回收率为3.87%。The method of filtration-centrifugal to extract hyaluronidase from bovine testicular was studied. The effects of extractant, holding time, separation method, salting reagent and the best dosage of salting reagent on activity of hyaluronidase were studied. The results showed the best reaction conditions:pH value of salt solution 3.5 (0.1 mol/L acetic acid + 1 mmol/L HCI +0. 1 mol/L NaCl)as the best selection of extraction solvent. After standing 8 h at 0 ℃, extraction solvent was separated with the method of filtration and centrifugal at 3 000 r/min. The highest enzyme activity was 277.5 U/mL, specific activity was 12.4 U/μg and the yield was 3.87% with ammonium sulfate 240 g/L as salting reagent.
