摘　　要：目的:以中医整体观指导,应用灌流法制作猫局灶脑缺血模型,用以观察未缺血脑区功能状态对缺血脑区损伤程度的影响。方法:猫麻醉后,以切割电锯开颅,充分暴露外斯氏前回皮层及其皮层支动脉(来源于大脑中动脉),经外斯氏前回皮层支动脉行穿刺灌流;5min后,取灌流区脑组织测定ATP含量。假手术组只暴露外斯氏前回皮层支动脉,不穿刺灌流。结果:灌流区在5s内即出现明显缺血变化。假手术组(n=6)与灌流缺血组(n=5)ATP含量分别为(1.28±0.23)pmol/gprot、(0.29±0.19)pmol/gprot,灌流缺血区ATP含量下降幅度达到77.3%,P<0.05。结论:灌流法制作猫局灶脑缺血模型制作成功,适用于研究未缺血脑区功能状态对缺血脑区损伤程度的影响。Objective： Under the guidance of holism in TCM,preparing focal cerebral ischemic model in cat by perfusing cortical artery with Ringer＇s solution in order to investigate the influence of functional status in non-ischemic cerebral region on the degree of injury in ischemic region.Methods： Anaesthesia of cats（2.5±3.5）kg was induced with 4% pentobarbital sodium in a dose of 40mg/kg（i.p）.Then,craniotomy was performed by dissection with electric saw,and ectosylvian gyrus and cortical artery to it,from the middle cerebral artery,were exposed.Cortical artery of ectosylvian gyrus was perfused with Ringer＇s solution,and proximal part of the artery was clipped.After perfusing for 5 min,the perfused cortical region was removed.ATP content of it was determined later.In fales-operated group,cortical artery was not perfused after exposed.Results： In 5 min,the perfused cortical region showed ischemia obviously.ATP contents of false-operated group and ischemic group induced by perfusing Ringer＇s solution were（1.28±0.23）pmol/gprot（n=6） and（0.29 ±0.19） pmol/gprot（n=5）,respectively,（P〈0.05）.The reduction of ATP in perfused region was 77.3%.Conclusion： Perfusion of cortical artery can be used to produce focal cerebral ischemia in cat.This model is suitable to investigate the influence of functional status in non-ischemic cerebral region on the degree of injury in ischemic region.
