益气活血中药复方与CVB3对乳鼠心肌细胞基因表达的影响

摘　　要：目的:克隆和筛选CVB3乳鼠心肌细胞感染相关基因,以期从基因水平揭示CVB3心肌炎的发病机制,同时探索益气活血中药复方治疗病毒性心肌炎的作用机制。方法:通过改良的抑制性消减杂交技术(suppression subtractive hybridization,SSH),分离大鼠心肌细胞CVB3感染组和益气活血中药复方给药组差异表达的基因,并通过荧光RT-PCR对上述结果进行验证。结果:益气活血中药复方治疗组ARMET(arginine-rich,mutated in early stage tumors)基因的表达比CVB3病毒感染组低,而PI3K样家族蛋白(Phosphoinositide 3-kinase-like family protein)催化区基因在益气活血中药复方治疗组表达则增加,并得到荧光RT-PCR的验证。结论:筛选到两个可能与CVB3心肌炎形成有关的基因,提示了病毒性心肌炎发生的新机制;益气活血法可能通过调节某些基因的表达而实现治疗病毒性心肌炎的目的。Objective： Clone and screening CVB3 lactation rats myocardial cell infection related genes from gene level,in order to reveal the mechanism CVB3 myocarditis,and seeking a qi and promoting blood circulation traditional Chinese medicine（TCM） and treatment of viral myocarditis mechanism.Methods： through the improvement of hybrid technology（suppression inhibitory cut subtractive hybridization,SSH）,separate rat heart muscle cells CVB3 infection group and qi and promoting blood circulation to traditional Chinese medicine（TCM） medicine group of genes differentially expressed,and through the fluorescence RT-polymerase chain reaction（PCR） of the above results were verified.Results： the qi and promoting blood circulation traditional Chinese medicine（TCM） and the treatment group ARMET arginine（flowers,mutated in stage care tumors） the expression of genes than CVB3 virus infection group low,and PI3K sample Phosphoinositide 3-（family protein kinase-like protein catalytic area genes don） in qi and promoting blood circulation traditional Chinese medicine（TCM） and the treatment group expression then increase,and get the fluorescence RT-PCR validation.Conclusion： screening to the two may be and CVB3 myocarditis form of the genes,tip the viral myocarditis new happened;Qi and promoting blood circulation laws can adjust the some the expression of genes and realize the aim of treatment of viral myocarditis.
