摘  要:目的:研究二烯丙基硫醚(Diallyl sulfide,DAS)对大鼠局灶性脑缺血再灌注损伤的保护作用和治疗时间窗。方法:DAS的保护作用研究,采用线栓法制备大鼠大脑中动脉阻断(Middle cerebral artery occlusion,MCAO)局灶性脑缺血模型,分别在造模前7 d采用腹腔注射法连续给予DAS 25、50、100、200 mg/kg,制备MCAO模型,缺血2 h,再灌注24 h,评价大鼠神经功能状态、脑水肿、脑梗死面积;DAS的治疗时间窗研究,采用MCAO局灶性脑缺血再灌注模型,分别在缺血1 h、再灌注损伤后2、4、6h腹腔注射DAS 50 mg/kg,缺血2 h,再灌注24 h,评价神经功能状态、脑水肿和脑梗死面积模型。结果:DAS 50 mg/kg,能最大程度的改善大鼠脑缺血再灌注后神经功能评分,减轻脑水肿和减少脑梗死面积;缺血后1 h开始用药,神经功能缺损评分明显减少,脑水肿及脑梗死面积也明显下降,再灌注2、4 h后给药,疗效下降,6 h给药,则无明显作用。结论:DAS对大鼠局灶性脑缺血再灌注损伤有保护作用,有效时间窗在缺血后1 h到再灌注损伤后2 h内。Objective:To investigate the protective effect of diallyl sulfide and the therapeutic window of opportunity for diallyl sulfide following focal cerebral ischemia/reperfusion injury in rats.Methods:For the protective effect of diallyl sulfide,focal cerebral ischemia-/reperfusion models in rats were induced by the transient occlusion of the middle cerebral artery(MCAO)for 2 h,followed by reperfusion for 24 h,and the animals were administered DAS25,50,100,200 mg/kg,ip,q d × 7 d before transient MCAO.The neurological deficit score,infarct size and brain edema were examined.For the therapeutic window of opportunity,the MCAO model was induced by transient occlusion for 2 h and reperfusion for 24 h.The drugs were administered at 1h after the onset of ischemia and 2,4,6 h after reperfusion respectively.The neurological deficit score,infarct size and brain edema were examined.Results:DAS 50 mg/kg,ip,q d × 7 d before MCAO significantly reduced the neurological deficit score,infarct size and brain edema.The administration of DAS beginning at 1 h after the onset of ischemia significantly reduced neurological deficit sore,infarct size and brain ischemia;starting at 2,4 h after reperfusion,the neuroprotective effects decreased,and starting at 6 h after reperfusion did not result in significant reduction of the above indexes for brain tissue.Conclusion:Diallyl sulfide has protective effect on cerebral ischemia/reperfusion injury.The therapeutic window of opportunity of diallyl sulfide in transient focal cerebral ischemia injury is between 1 h after the onset of ischemia and 2 h after reperfusion.
