摘 要:目的:探索Snail基因与食管癌细胞侵袭迁移能力的关系。方法:体外合成Snail序列特异性双链小干扰RNA,转染人食管癌细胞EC9706,应用RT-PCR及Western blotting方法检测干扰效果,细胞划痕实验检测细胞迁移能力变化,Transwell小室侵袭实验检测细胞侵袭能力变化。结果:经Snail特异性siRNA干扰后细胞Snail基因及蛋白表达水平明显降低,细胞的侵袭迁移能力随之降低,差异均有统计学意义(均P<0.05)。结论:Snail基因与食管癌细胞的侵袭迁移能力有关,采用siRNA干扰Snail基因的表达能有效逆转食管癌细胞的侵袭迁移能力。Objective: To study the relativity between Snail gene and invasion and migration ability of esophageal carcinoma cells. Methods: The chemically synthetic siRNA duplexes targetting Snail was transiently transfected into EC9706 cells. After siRNA interfering cells were harvested, the expression of Snail was investigated by of RT-PCR and Western blotting. Migration rate was measured by wound healing assay. Furthermore, the ability of cell invasion was investigated using Transwell chambers assay. Results: Snail-siRNA effectively inhibited the expression of Snail in gene and protein level and the ability of cell invasion and migration decreased either, the difference was statistically significant (all P〈0.05). Conclusion: Snail contribute to the invasiveness of esophageal carcinoma cell, siRNA interference the expression of Snail gene can reverse invasion and migration ability of esophageal carcinoma cells effectively.
