摘 要:目的探讨STAT5、SOCS1和PTPRO基因在急性白血病(AL)中的表达情况及临床意义。方法将入选患者分为AL初治组、完全缓解组(CR组)和正常对照组(NC组),Syber Green荧光定量PCR方法检测各组骨髓单个核细胞(MNC)中STAT5、SOCS1及PTPRO mRNA的表达。结果 (1)AL初治组STAT5 mRNA表达水平明显高于缓解组(P<0.05),缓解组STAT5表达水平高于正常对照组(P<0.05);(2)AL初治组SOCS1、PTPRO的表达水平低于缓解组(P<0.05),缓解组SOCS1、PTPRO的表达水平低于正常对照组(P<0.05);(3)STAT5、SOCS1、PTPRO基因表达水平中位值分别为0.980、0.838和0.771,根据中位值分为高表达组和低表达组,两组缓解率间差异无统计学意义(P>0.05)。结论 STAT5的高表达及SOCS1和PTPRO的低表达可能在白血病的发生、发展中起重要作用。Objective To investigate the expression levels and the clinical significance of STAT5,SCOS1 and PTPRO genes in leukemia patients.Methods The enrolled subjects were divided into three groups including the de novo AL group,the complete remission(CR) group and the normal control(NC) group(comprised by healthy volunteers).Quantitative Syber Green polymerase chain reaction(PCR) was applied to detect the STAT5,SOCS1 and PTPRO mRNA levels in marrow mononuclear cells(MNC) of all the three groups.Results STAT5 mRNA expression was significantly higher in the de novo AL group than that in the CR group(P0.05),and higher in the CR group than that in the NC group(P0.05).SOCS1 and PTPRO mRNA expression was lower in the de novo AL group than that in the CR group(P0.05) and lower in the CR group than that in the NC group(P0.05).All subjects were divided into the higher expression group and the lower expression group according to the median expression levels of STAT5,SOCS1,and PTPRO genes,0.980,0.838 and 0.771 respectively.However,no significant differences were observed in remission rate between the high expression groups and the low expression groups.Conclusion Higher STAT5 expression and lower SOCS1 and PTPRO expression may have played important roles in the pathogenesis and development of acute leukemia.
