参芎化瘀胶囊对大鼠全脑缺血损伤的保护作用

摘  要:目的探讨参芎化瘀胶囊对缺血性脑卒中的治疗作用及机制。方法 SD大鼠分为假手术组、模型组、参芎化瘀胶囊高、低剂量组。改良的Pulsineli 4血管阻断(4-VO)法全制作脑缺血模型。免疫组织化学法检测海马区β淀粉样肽(Aβ)表达,TUNEL法检测凋亡细胞,八臂迷宫法测试动物学习记忆功能。结果与对照组比较,模型组海马区Aβ蛋白、凋亡神经细胞数量增加;大鼠的习记忆功能下降;与模型组比较,参芎化瘀胶囊组中海马区Aβ表达,神经细胞凋亡数量回降;同时动物学习记忆功能改善,上述变化在高剂量参芎化瘀胶囊组更为明显。结论参芎化瘀胶囊对脑缺血损伤有很好的治疗作用,其机制与抑制Aβ沉积有关、神经细胞凋亡有关。Objective To investigate the effects of Shenxionghuayu capsule on cerebral ischemia and its underlying mechanism in rats.Methods Male SD rats were divided randomly into 4 groups: sham,model,low and high dose of Shenxionghuayu capsule treatment groups.Global cerebral ischemia model was formed by improved four-vessel occlusion as described according to the description of Pulsinelli′s method.The expression of Aβ was detected by immunohistochemistry.The quantities of apoptotic cells was measured by TUNEL method.Learning and memory functions were performed with eight-arm maze.Results Results Compared with sham group,the Aβ expression and the number of neural apoptotic cells were enhanced and learning and memory functions were significantly decreased in model group.Compared with model group,the Aβ expression and the number of neural apoptotic cells were decreased obviously,learning and memory functions significantly were improved in Shenxionghuayu capsule groups.Changes of above indexes were more significant in high dose of Shenxionghuayu capsule group.Conclusions Shenxionghuayu capsule has good therapeutic effect on cerebral ischemia injury,which is related to regulating Aβ expression and reducing neuronal apoptosis.
