非甾体类抗炎药NAG-1在胃癌中表达及意义

摘　　要：目的研究非甾体类抗炎药活化基因-1(NAG-1)在胃癌组织中的表达及其与临床病理特征的关系。方法采用RT-PCR和免疫组化检测76例胃癌组织及其相应正常组织NAG-1的表达。结果胃癌组织的NAG-1蛋白的表达程度显著低于正常组织(χ2=47.186,P<0.05)。NAG-1蛋白在胃癌组织中的表达与患者的细胞分化程度有关(χ2=7.506,P<0.05)。而与性别、淋巴结转移、临床分期间的差异无统计学意义(P>0.05)。RT-PCR结果显示NAG-1 mRNA在胃癌组织中的相对含量显著低于正常胃黏膜组织。NAG-1 mRNA在高、中分化胃癌中的相对含量显著高于低、未分化(P<0.05)。而与淋巴结转移、临床分期、浸润深度差异无统计学意义(P>0.05)。结论 NAG-1基因在胃癌组织中的表达下降,可能参与胃癌的发生、发展过程。Objective To study the expression of nonsteroidal anti inflammatory drug - activated gene 1 in human gastric cancer and its relationship with the clinical staging,differentiation degree,and lymphatic metastasis of gastric cancer. Methods The ex pression of NAG-1 was detected by semi-quantitative reverse transcriptase polymerase chain Reaction （RT PCR） and immunohistochemistry（IHC） in the cancer tissues and corresponding adjacent normal tissues in 76 gastric cancer patients. Results Immuno-histochemistry showed the expression of NAG-1 protein in the cancer tissues was lower than that in the normal tissues （x^2 =47. 186, P〈0.05） ;and the expression was relationship with differentiation degree（x^2 = 7. 506, P^0.05）, but the expression was not associ ated with the depth of tumor invasion,lymph node metastasis,and clinical stage（all P〉0.05）. The result of RT-PCR showed that NAG-1 mRNA expression in gastric cancer tissue significantly was lower than that in normal tissues（P〈0.05）. NAG-lmRNA ex- pression in well and moderately differentiated cancer cells was significantly higher than those of poorly and non-differerntiated cancer cells（P〈0.05）, the expression was not associated with the depth of tumor invasion, lymph node metastasis, and clinical stage（P〉0.05）. Conclusion Decreased expression of NAG-1 in human gastric cancer,and it may play an important role in the tu morigenesis of gastric cancer.
