摘  要:目的研究人脂联素重组体(RA)对人子宫内膜癌细胞系RL95-2体外侵袭能力的影响,探讨RA治疗子宫内膜癌临床应用的可行性。方法将RL95-2细胞分设对照组和RA加药组,作用一定时间后,分别以MTT法、黏附实验、Transwell小室实验、体外细胞迁移实验检测RA对人子宫内膜癌细胞增殖、黏附、侵袭和迁移能力的影响。结果 RA对ECM和FN黏附能力影响的程度不同,对ECM的敏感性大于FN。RA浓度高于5.0μg/ml时,对RL95-2细胞生长具有明显的抑制作用(P<0.01),并能明显地降低肿瘤细胞体外侵袭能力((P<0.01)),侵袭抑制率随RA浓度的升高而增加。RA浓度高于5.0μg/ml时,明显地抑制肿瘤细胞的迁移能力(P<0.01)。结论脂联素重组体在大于5.0μg/ml时,可抑制人子宫内膜癌细胞系RL95-2的侵袭及转移能力,RA可望成为晚期、无法耐受手术和大剂量放化疗及复发性子宫内膜癌患者的综合治疗方法之一。Objective To investigate the effects of recombinant adiponectin(RA) on the invasive ability of human ndometrial carcinoma cell line RL95 - 2 and explore the feasibility of clinical application of RA in the treatment of endometrial carcinoma. Methods RL95 - 2 ceils were divided into control group and RA(2. 5,5.0,10,20, and 30μg/ml)treatment groups. The effect of RA on proliferation of RL95 -2 cells was measured by MTT assay, cell adhesion assay was carried out in a microcuhure well precoated with fibronectin, the invasive ability of RL95 - 2 cell were assayed in Transwell cell culture chamber, and migration ability were observed by cell wound healing assay. Results RA was more sensitive to ECM and than to FN. The growth and invasive ability of RL95 -2 cells were obviously inhibited when the concentration of RA was higher than 5.0 μg/ml (P 〈0. 01 ) ,the inhibition of migration was in a concentration - dependent manner with the inhibitory rate. Conclusion These results indicate that adiponectin is effective in suppressing tunor cell invasion, and adiponectin may be one of the combined modality therapies for the patients in advanced stage or being too weak to tolerate surgery and large dose radiotherapy and chemotherapeutic drug or recurrent endometrial carcinoma.
