超声辅助萃取HPLC法测定烟草中蔗糖、果糖、葡萄糖含量

摘　　要：[目的]建立超声辅助萃取HPLC法测定烟草中蔗糖、果糖、葡萄糖含量的方法。[方法]样品采用水提取,超声萃取,C18固相萃取柱净化,以乙腈∶水=70∶30（V/V）为流动相,经Agilent Zorbaxcarbohydrate柱（4.6 mm×250.0 mm,5μm）检测,流速为1.0 ml/min,柱温为30℃,示差折光检测器温度为35℃。[结果]该法RSD为0.89%-0.98%（n=5）,加标回收率为98.02%-99.16%,糖类质量浓度在0.625-20.000 mg/ml内与峰面积呈现良好的线性关系,相关系数在0.998 5（n=6）以上。[结论]该方法快速准确,是检测烟草蔗糖、果糖、葡萄糖含量的可靠方法。[Objective] The aim was to set up methods for the determination of Sucrose,fructose,glucose in tobacco.[Method] Sampling water was used as solvent,then extracted by ultrasonic,coupled with solid-phase extraction.C18 solid phase column was purified and acetonitrile： water=70∶30（V/V） as moving phase.The chromatographic separation was achieved on an Agilent Zorbaxcarbohydrate column（4.6 mm× 250mm,5 μm）,which was held at a constant temperature of 30 ℃,using a mobile phase composed of acetonitrile and water（70∶30,V/V） at a flow rate of 1.0 ml/min.RID temperature was set as 35 ℃.[Result] Average spike recoveries for 5 replicate determinations of three saccharides were found in a range of 98.02% to 99.16%,with a relative standard deviation（RSD） between 0.89% and 0.98%.The developed calibration curves displayed good linearity over a concentration range of 0.625 to 20 mg/ml,with a correlation coefficient exceeding 0.998 5（n=6）.[Conclusion] The proposed method was accurate and was dependable method for the analysis of these sugars in tobacco.
