摘 要:目的:观察缺血后处理对局灶性脑缺血再灌注大鼠SOD、MDA的影响。方法:线栓法建立大鼠大脑中动脉缺血再灌注模型,将成年雄性Wistar大鼠144只,随机分成三组:假手术组、对照组(脑缺血再灌注组)、缺血后处理组。假手术组只进行假手术;对照组(脑缺血再灌注组)给予90 min大脑中动脉缺血(MCAO);缺血后处理组在大脑中动脉缺血90 min后,给予灌注30 s缺血30 s,反复3次。各组分别再灌注12 h、24 h、48 h、72 h后测定脑组织超氧化物歧化酶(SOD)、丙二醛(MDA)水平的改变。结果:局灶性脑缺血后再灌注前给予后处理,缺血后处理组与缺血再灌注组相比,SOD的表达明显增加,而MDA的表达明显减少。结论:脑缺血后处理增加脑缺血再灌注后SOD的表达、减少MDA的表达,进而提高脑组织的抗氧化能力。Objective:To investigate the effect of focal cerebral isehemic postconditioning on the expression of SOD, MDA. Methods:The model of focal brain ischemia was made by an intraluminal suture occulusion of the middle cerebral artel7 ( MCA ) in rats. 144 aduh male wistar rats were divided randomly into 3 groups:sham - operated group,controlled group( ischemia -reperfusion group) ,isehemic postconditioning group. Sham -operated group received sham surgery only. And the controlled group received 90 minutes of middle cerebral artery occlusion(MCAO) . But the ischemic postcon- ditioning group received the three cycles of 30 sees of reperfusion and 30 sees of middle cerebral artely occlusion after 90 minutes of middle cerebral artery occlusion (MCAO). After all groups were subjected 12 ,24 ,48 and 72 hours of the reperfusion, the activities of SOD and the concentration of MDA were measured in every group. Results:The expression of SOD in isehemic postconditioning group were obviously higher than it in isehemia - reperfusion group at different reperfusion time points,and the expression of MDA in ischemic postconditioning group were lower than it in ischemia -reperfusion group at different reperfusion time points. Conclusions: Ischemie postconditioning increase the expression of SOD and reduce the expression of MDA to enhance the anti - oxidative ability in brain after cerebral - schemia/ reperfusion.
