信号转导与转录因子3及其磷酸化在宫颈癌组织中的表达及其临床意义

摘　　要：目的:探讨信号转导与转录因子3(STAT3)在不同病理类型宫颈癌中的表达及其临床意义。方法:采用免疫组化法检测15例正常宫颈组织和40例宫颈癌组织中STAT3的表达,采用Western blot法检测40例宫颈癌组织中STAT3及磷酸化STAT3(p-STAT3)的表达。结果:(1)免疫组织化学染色显示,在宫颈癌组织中STAT3表达升高;(2)宫颈癌组织中STAT3蛋白表达水平和磷酸化水平明显高于对照组(P<0.05);(3)宫颈癌组中鳞癌的STAT3蛋白表达率和磷酸化水平最高,腺鳞癌其次,腺癌最低,但三者比较差异均无统计学意义(P>0.05)。结论:STAT3及其磷酸化蛋白在宫颈癌组织中呈现高表达,有望成为宫颈癌判断预后和靶向治疗的目标。Objcetive：To investigate the expression of Signal Transducer and Activator of Transcription 3（STAT3）in the cervical cancer.Methods：The expression of STAT3 protein was detected by immunohistochemistry（SP method） in 15 healthy cervix tissues and 40 cervix cancer tissues.Protein was extracted and protein levels of unphosphorylated and phosphorylated STAT3 were measured by Western blot.Results：The positive expression of STAT3 in cervix cancer was showed.levels of unphosphorylated and phosphorylated STAT3 in cervix cancer were higher than normal tissue.The level of unphosphorylated and phosphorylated STAT3 in squamour cervix cancer was the most,but no significant differences was showed.Conclusion：High level of unphosphorylated and phosphorylated STAT3 plays an important role in cervix cancer.STAT3 may be regard as a noble marker to evaluate prognostic and curative effect of cervix cancer.
