高效液相色谱法测定跌打活血散中儿茶素和表儿茶素的含量

摘　　要：目的建立跌打活血散中儿茶素和表儿茶素的含量测定方法。方法高效液相色谱法,采用Prevail Select C18(4.6mm×150 mm,5μm)色谱柱,柱温35℃;以乙腈-水-磷酸(100∶900∶1)为流动相;流速:1 mL.min-1;检测波长278 nm。结果儿茶素线性范围为30.08～150.4μg.mL-1,r=0.999 9;表儿茶素线性范围为12.24～61.20μg.mL-1,r=0.999 9;儿茶素和表儿茶素的回收率分别为98.4%和96.5%,RSD分别为0.92%和1.3%。结论本方法操作简便,测定结果准确可靠,可用于跌打活血散中儿茶素和表儿茶素的含量测定。OBJECTIVE To establish a method for determining catechin and epicatechin in Dieda Huoxue San.METHODS High performance liquid chromatography was used for determining the contents of catechin and epicatechin.The chromatography column was Prevail Select C18（4.6 mm×150 mm,5 μm）and the column temperature was 35 ℃.The mobile phase was acetonitrile-water-phosphoric acid（100∶900∶1）with a flow rate of 1 mL·min-1.The detection wavelength was 278 nm.RESULTS The linear range of catechin was 30.08-150.4 μg·mL-1（r=0.999 9）.The average recovery of catechin was 98.4% and RSD was 0.92%.The linear range of epicatechin was 12.24-61.20 μg·mL-1（r=0.999 9）.The average recovery of epicatechin was 96.5% and RSD was 1.3%.CONCLUSION This method is simple,time-saving and accurate.It can be used for routine analysis of catechin and epicatechin in Dieda Huoxue San.
