摘  要:目的:探讨血浆同型半胱氨酸(Hcy)水平与大鼠学习记忆功能之间的关系以及叶酸干预对改善学习记忆能力的效果。方法:24只SD大鼠随机分为正常组、蛋氨酸组、蛋氨酸+叶酸组,正常组给予普通饲料喂养,对蛋氨酸组及蛋氨酸+叶酸组在饲料中添加3%的蛋氨酸制备高同型半胱氨酸血症模型,同时正常组、蛋氨酸组给予0.8 mg(/kg.bw)的生理盐水灌胃,蛋氨酸+叶酸组给予灌胃0.8 mg(/kg.bw)的叶酸。ELISA试剂盒检测血浆同型半胱氨酸、Morris水迷宫检测大鼠学习记忆能力。结果:蛋氨酸组血浆Hcy水平高于正常组,蛋氨酸+叶酸组血浆Hcy含量低于蛋氨酸组(P<0.05);定位航行实验第3天、第4天,蛋氨酸组逃避潜伏期时间高于正常组,蛋氨酸+叶酸组低于蛋氨酸组(P<0.05);空间探索实验,蛋氨酸组大鼠在第一象限停留时间少于正常组,路径长度高于正常组(P<0.05)。蛋氨酸+叶酸组在第一象限时间长于蛋氨酸组,路径长度短于蛋氨酸组(P<0.05)。结论:血浆Hcy水平的升高影响大鼠学习记忆能力,通过叶酸补充可以降低血浆Hcy水平,改善大鼠学习记忆能力。Objective: To explore the relationship between plasma homocysteine and learning and memory ability of rats and observe the effects of folate on these abilities. Methods: 24 SD rats were divided randomly into three groups,which were normal group, methionine group and methionine + folate group. The normal group were given normal diet. Groups of methionine and methionine + folic acid were added methionine( 3%)in rat feed to establish hyperhomocysteine(HHcy) model. While the normal group and methionine group were received 0.8 mg/(kg·bw) of normal saline via intragastric intubation, and methionine + folate group were given 0.8 mg/(kg·bw) of folic acid. Plasma homocysteine was detected by ELISA Kit. Learning and memory abilities were measured by Morris water maze. Results: Plasma homocysteine level in methionine group was higher than that in normal group (P〈0.05). Plasma homoeysteine level in methionine + folate group was lower than that in methionine group (P〈0.05). In place navigation test, escape incubation period of time in methionine group was longer than that in normal group. Same indicator in methionine + folate group was shorter than that in methionine group (P〈0.05). In spatial probe test, the first quadrant retention time in methionine group rats was shorter than that in normal group. Same index in methionine + folate group was longer than that in methionine group. Path length of those 3 groups showed opposite trend(P〈0.05 ). Conclusion: Learning and memory ability is decreased by elevated plasma homocysteine in rats. Folate supplementation can reduce plasma homocysteine levels and improve the ability of learning and memory. Key words
