摘 要:目的观察阿托伐他汀对糖尿病ApoE基因敲除小鼠主动脉组织中CD55、CD59表达的影响。方法 22只雄性6周龄ApoE基因敲除小鼠随机分为糖尿病动脉粥样硬化(对照)组(12只)和阿托伐他汀干预(干预)组(10只),予链脲佐菌素(55 mg.kg-1.d-1体质量)连续5次腹腔注射建立糖尿病模型,普通饲料喂养。造模成功后,干预组予阿托伐他汀(10 mg.kg-1.d-1体质量)灌胃8周。监测生化指标,测量斑块面积;采用免疫荧光和Real-time PCR技术,观察阿托伐他汀干预后小鼠主动脉组织中CD55和CD59的表达变化。结果与对照组相比,阿托伐他汀干预后体质量增加,血糖和血脂无变化,血清淀粉样蛋白-A[(5.9±3.2)μg/mL vs.(3.8±1.6)μg/mL,P>0.05]和动脉粥样硬化病变面积呈减少趋势[(264046.4±88374.97)μm2vs.(218752.4±87927.16)μm2,P>0.05]。免疫荧光显示,阿托伐他汀干预后小鼠主动脉组织中CD55和CD59表达较对照组增强。Real-time PCR结果显示,与对照组比较,阿托伐他汀干预后CD55和CD59 mRNA相对表达量无明显变化(0.55±0.2 vs.0.43±0.11,0.76±0.36 vs.0.55±0.13,P均>0.05)。结论独立于调脂作用之外,阿托伐他汀可能具有上调CD55和CD59蛋白表达的作用。Objective To investigate the effects of atorvastatin on CD55 and CD59 expressions on aortas in streptozotocin-induced diabetic ApoE-knock out mice.Methods At 6 weeks of age,twenty-two male ApoE-knock out mice were obtained and divided into two groups: diabetic atherosclerotic group(n=12),and atorvastatin group(n=10).Mice were rendered diabetic by intraperitoneal injection of streptozotocin 55 mg/kg of body weight/day for 5 consecutive days.After 8 weeks of treatment with atorvastatin 10 mg/kg of body weight/day,venous bloods were collected for glucose and lipids analysis,and aorta were prepared for atherosclerotic plaques,immunofluorescence and Real-time PCR.Results There were no differences in glucose and lipids between diabetic atherosclerotic group and atorvastatin group.After treatment with atorvastatin,there was a trend towards decrease in serum amyloid-A[(5.9±3.2)μg/mL vs.(3.8±1.6)μg/mL,P0.05)]and the atherosclerotic plaques[(218752.4±87927.16)μm2 vs.(264046.4±88374.97)μm2,P0.05)].Immunofluorescence showed that CD55 and CD59 protein expressions increased in atorvastatin group compared with diabetic atherosclerotic group.Real-time PCR showed that levels of CD55 and CD59 mRNA were similar in diabetic atherosclerotic group and in atorvastatin group(0.55±0.2 vs.0.43±0.11,0.76±0.36 vs.0.55±0.13,P0.05).Conclusion Independent of the effects on lipids,atorvastatin could increase CD55 and CD59 protein expressions in diabetic ApoE-knock out mice.
