摘 要:目的应用细胞微囊化方法、病理技术研究微囊化大鼠胰岛细胞与睾丸Sertoli细胞联合移植对糖尿病小鼠的血糖影响及Sertoli细胞对共移植的胰岛细胞的免疫豁免作用。方法将30只小鼠随机分为2组,每组15只。Ⅰ组为胰岛细胞微囊化移植组,Ⅱ组为胰岛细胞与睾丸Sertoli细胞联合微囊化移植组,建立糖尿病模型后,2组均采用腹腔注射的方法将微囊植入糖尿病小鼠体内,术后2组均不使用免疫抑制剂,定期检测小鼠的血糖情况,比较2组之间胰岛细胞有功能存活时间的不同以及微囊的病理学改变。结果胰岛细胞与睾丸Sertoli细胞联合微囊化移植组与胰岛细胞微囊化移植组相比,联合移植组胰岛细胞存活时间长于单纯移植组(P<0.05),囊周淋巴细胞浸润程度轻于单纯胰岛细胞移植组。结论睾丸Sertoli细胞对共移植的胰岛细胞具有免疫豁免作用,可以延长胰岛细胞的有功能存活时间,减轻排斥反应,降低囊周淋巴细胞浸润程度。Objective To investigate the effect of combined transplantation of microencapsulation pancreatic islet with testicular Sertoli cell on blood glucose in rats with diabetes mellitus and the effect of Sertoli cell on immune exemption for co - transplanted islet ceils through cell microencapsulation method and pathological technique. Methods Thirty mice were randomly divided into two groups, simple islet cell microencapsulation transplantation group ( n = 15) and combined transplantation group ( n = 15). After the diabetic models of mice were established,the microcapsule was implanted into the body of diabetic mice by peritoneal injection for both groups, and immunosuppressive agents were not administrated for both groups, then blood glucose was detected, the survival time of islet cells and the pathological changes of microcapsule were observed and compared between the two groups. Results The survival time of islet cells in combined transplantation group was significantly longer than that of simple islet cell microencapsulation transplantation group ( P 〈 0.05 ), however, the infiltration degree of lymphocytes around the microcapsule in combined transplantation group was obviously slighter than that in simple islet cell microencapsulation transplantation group. Conclusion The testicular Sertoli cells have immune exemption effect for co-transplanted islet cells, which can prolong the survival time of functional islet ceils, relieve rejection reaction and decrease the infiltration extent of lymphocyte around the microcapsule.
