摘 要:目的研究外源性血管内皮生长因子(VEGF)局部应用对大鼠损伤脊髓神经组织的作用。方法采用改良Allen氏重量打击法大鼠急性脊髓损伤模型。脊髓打击性损伤后,将模型大鼠随机分组,并依组别在蛛网膜下腔分别给药。A组:生理盐水10μL,B组:Matrigel蛋白胶10μL,C组:重组人VEGF_(165) 0.2μg+Matrigel蛋白胶10μL。脊髓损伤后1、2、3、4周对损伤脊髓组织进行病理组织学研究,观察脊髓灰质神经元数量。结果在损伤后1~4周,VEGF治疗组动物脊髓损伤区残留组织多,残留组织增加。脊髓灰质神经元数量明显优于同期对照组。结论外源性VEGF局部应用,能够保护大鼠损伤后的脊髓组织。Objective To investigate the protective effect of exogenous vascular endothelial growth factor (VEGF) on spinal cord tissue after spinal cord injury (SCI) in adult rats. Methods A contusion injury was produced to adult rats by using the modulated Allen's weight drop method. After SCI, seventy-two model rats were randomly divided into three groups, and the drugs were injected into subrachnoid space at the lesion site. In group A: saline 10 μL, in group B: Matrigel 10 μL, in group C: human recombinant VEGF165+Matrigel 10 μL. 1, 2,3, 4 weeks after surgery, animals were killed to study spinal cord pathologic histology and the number of neurons. Results VEGF-treated animals had an increased amount of spared tissue in the lesion area. The number of neurons in VEGF-treated animals was much better than that in the control groups at all time points following SCI. Conclusion The exogenous vascular endothelial growth factor can protect spinal cord tissue from damage caused by spinal cord injury.
