摘 要:目的探讨Sonichedgehog(SHH)在肝细胞癌(HCC)和癌周肝组织中的表达及其临床意义。方法检测145例HCC和癌周肝组织SHH的表达,了解其表达与患者根治性切除术后预后的关系。检测4种肝癌细胞株以及正常肝细胞株SHHmRNA的含量。结果SHH主要表达于肝细胞和肝癌细胞的胞质,肝癌与癌周肝组织表达差异无统计学意义(P〉0.05)。癌和癌周高表达的患者总体生存率显著高于低表达患者(P〈0.05)。肝癌细胞株SHH表达量显著高于正常肝细胞株(P〈0.01)。结论SHH在HCC和癌周肝组织中高表达,其表达可用于区分患者预后,SHH可能参与HCC的发生和发展。Objective To investigate the expression of Sonic hedgehog (SHH) in paired tumor and peritumoral liver tissues of hepatoeellular carcinoma ( HCC ) and its clinical significance. Methods The expression of SHH was assessed by immunohistoehemistry in tissue microarrays containing paired tumor and peritumoral liver tissues from 145 patients who received hepateetomy for histologically proven HCC. Prognostic value of SHH expression was evaluated. The SHH mRNA expression of four HCC cell lines (HCCLM3, MHCC97H, MHCC97L, HepG2) and a normal liver cell line (L-02) was detected by realtime reverse transcription-polymerase chain reaction (RT-PCR). Results SHH was mainly expressed in the cytoplasm of tumor cells or hepatocytes in peritumoral or normal liver tissues. There was no statistically significant difference in the SHH density betwen intratumoral and peritumoral liver tissues ( P 〉 0. 05 ). The median overall survival (OS) of HCC patients with high intratumoral SHH expression was longer than low SHH expression patients ( P 〈 0. 05 ). High peritumoral SHH expression was also associated with good survival ( P 〈 0. 05 ). SHH was expressed positively in both HCC and normal cell lines, and the levels of mRNA was higher in HCC cell lines than in L-02 ( P 〈 0. 01 ). Conclusion SHH is expressed higher in both intratmnoral or peritumoral liver tissues, and the expression of SHH is significantly correlated with OS of patients with HCC. SHH may become a biomarker of HCC patients after curative resection.
