摘 要:目的观察47℃不完全热消融后肝癌细胞对内皮细胞功能的影响。方法以47℃恒温水浴处理HepG2细胞;收集HepG2母系和亚系的条件培养基;检测条件培养基对人脐静脉内皮细胞(HUVECs)增殖、迁移和成管能力的影响。结果HepG2经47℃处理后产生快速增殖的亚系n;与母系比较,亚系n条件培养基显著促进HUVECs的增殖、迁移和成管,其分光光度值、迁移细胞数和成管环数明显增高,分别为0.57±0.04、85.60±9.40、18.00±1.90(P〈0.05);应用贝伐珠单抗后差异无统计学意义(P〉0.05)。结论肝癌细胞不完全热消融后可转化为快速增殖的亚系,后者可通过增加血管内皮生长因子(VEGF)的分泌增强内皮细胞的功能。Objective To investigate the effect of hepatocellular carcinoma (HCC) cells on the function of endothelial cells after 47 ℃ induced insufficient thermal ablation and the action mechanism. Methods HepG2 cells were treated with a 47℃ water bath. The conditioned media of the parental HepG2 and the subline cells were collected. Proliferation, migration and tube formation of human umbilical vein endothelial cells (HUVECs) were measured after the treatment with conditioned media. Results The treatment with a water bath of 47 ℃ resulted in transformation of HepG2 cells into the HepG2 n subline with significantly higher proliferation rate. After the treatment with conditioned media from the subline, the ahsorbance of HUVECs proliferation, number of cell migration and tube number were0. 57 ± 0. 04, 85.60 ± 9.40 and 18. 00 ± 1.90 respectively, which were significiantly higher than the parental one (P 〈 0. 05 ). Furthermore, bevaeizumab could eliminate the significant difference ( P 〉 0. 05 ). Conclusion Insufficient radiofrequency ablation can transform HCC cells into a subline with higher proliferation rate, and the latter could increase the ability of HUVECs in proliferation, migration and tube formation through the VEGF-de- pendent mechanism.
