摘  要:目的探讨大鼠抑郁模型治疗前后行为学及海马MR体积的动态变化及其意义。方法80只行为学评分接近的SD大鼠按照随机数字表法分为对照组(10只)、抑郁组(10只)及治疗组(60只),治疗组再均分为A、B、C3组,分别于建模第4周、6周及8周进行治疗。治疗分为自然恢复和氟西汀腹腔注射2种方式。抑郁模型运用慢性不可预知应激及孤养相结合的模式建立,应激共8周。对照组不做任何处理。建模开始前及每两周末进行大鼠体重测量、液体消耗实验,并于实验开始前、开始后每两周末及治疗后进行Moms水迷宫实验及在高场强MR上测量每组大鼠各侧海马体积。结果各项行为学指标表明建模成功并呈动态变化。与对照组相比.建模第4周,抑郁组大鼠左侧海马体积明显减小,差异有统计学意义(P〈0.05),右侧无明显变化;第6周时,右侧也明显减小,与对照组比较差异有统计学意义(P〈0.05)。治疗后,在自然恢复方式下,行为学于前期好转,中后期不但没有恢复,反而恶化,而海马体积均没有明显变化;在氟西汀腹腔注射治疗方式下,行为学指标好转,但海马仅左侧在前期出现一定程度地恢复,中后期没有明显差异。结论大鼠抑郁模型的行为学及海马体积呈动态变化,治疗后其行为学及海马体积表现各自不同的恢复性。Objective To explore the dynamic changes ofhippocampal volumes by MRI and their behavioral changes in rat depression models before and after antidepressant treatment, and their significance. Methods Based on the evaluation of ethology, 80 SD rats were randomly divided into 3 groups: control group (n=10), depression group (n=10), and treatment group (n=60) which was equally divided into 3 subgroups (group A, B and C), given 2 different treatments as self-recovery or Fluoxetine intraperitoneal injection in the 4^th, 6^th and 8^th week. The depression models were established by chronic unpredictable mild stress or isolated feeding for 8 weeks. The body weight and fluid consumption were measured before the antidepressant treatment and every 2 weeks of the treatment, and Morris water maze test was performed and the bilateral hippocampal volumes by high field MRI were measured before and after the antidepressant treatment and every 2 weeks of the treatment. Results The behavioral data indicated that the rat depression models were established successfully and these indexes were changed dynamically. In the 4^th week, as compared with those of the control rats, the left hippocampal volumes of depression rats were significantly decreased (P〈0.05), while the right ones were not significantly different. In the 6^th week, as compared with those of the control rats, the fight ones began to significantly decrease the volume (P〈0.05), and the left ones kept decreasing. By self-recovery, the ethology changed better in the prophase but worsened in the mid-late phase, and the hippocampal volumes had no significant change. By Fluoxetine treatment, the ethology changed better, and the left hippocarnpal volumes had significant changes only in the prophase. Conclusion The behavior and hippocampal volumes in rat depression models change dynamically, and after treatment, the ethology and hippocampal volumes change differentially.
