摘 要:目的观察骨髓间充质干细胞(Mesenchymal stem cells,MSCs)移植对大鼠溃疡性结肠炎(Ulcerative colitis,UC)的治疗作用。方法分离、培养、鉴定大鼠MSCs,并以DAPI标记第3代MSCs。将大鼠随机分为3组,A、B组采用免疫复合法诱导UC模型,C组为正常对照组。A组移植标记的MSCs,B组移植PBS。移植后3、7、14 d各组均处死大鼠5只,取结肠组织,光镜观察病理变化,荧光显微镜观察标记细胞的分布,激光共聚焦显微镜观察标记细胞细胞角蛋白(Cytokeratin,CK)的表达。结果 MSCs生长迅速,呈均一的成纤维样细胞,表达CD29和CD90,不表达CD45。移植后7、14 d,A组结肠的修复明显优于B组;移植后3 d,A组结肠即可见移植细胞,至14 d时最多(P<0.05);移植后7、14 d,A组少量移植细胞表达CK。结论 MSCs移植可促进UC大鼠损伤结肠的修复;部分MSCs可分化为结肠上皮细胞,参与损伤的修复。Objective To observe the curative effect of transplanted bone marrow-derived mesenchymal stem cells(MSCs) on ulcerative colitis(UC) in rats.Methods Rat MSCs were isolated,cultured and identified,of which the passage 3 were labeled with DAPI.Rats were divided into three groups randomly.The rats in groups A and B were induced by immune-combined TNBS / ethanol to establish UC model,while those in group C were served as normal control.The rats in group A were transplanted with DAPI-labeled MSCs,while those in group B with PBS.Five rats in each group were killed 3,7 and 14 d after transplantation respectively,of which the colon tissues were observed for pathological change by light microscopy,for distribution of DAPI-labeled MSCs by fluorescent microscopy,and for expression of cytokeratin(CK) by laser confocal microscopy.Results MSCs grew rapidly and showed a homogeneous fibroblast-like morphology,in which CD29 and CD90 were expressed while CD45 were unexpressed.The reparation of injured colon tissues of rats in group A 7 and 14 d after transplantation was significantly superior to that in group B.Transplanted MSCs were observed in group A 3 d and reached a maximum in quantity 14 d after transplantation(P 0.05).CK was expressed in a small quantity of transplanted MSCs in group A 7 and 14 d after transplantation.Conclusion Transplanted MSCs promoted the reparation of injured colon tissues of rats with UC,a partial of which might be differentiated into colon epithelial cells and involve in the reparation of injured colon.
