摘 要:目的探讨小鼠胚胎早期造血发育阶段Notch1信号的表达。方法采用RT-PCR和Western blot法对小鼠胚胎9.5 d(E9.5)、E10.5、E11.5和E12.5的主动脉-性腺-中肾(Aorta-gonad-mesonephros,AGM)区和胎肝中Notch1信号基因和蛋白的表达水平进行检测。结果 AGM区Notch1基因mRNA的表达量较高,且随孕龄的增大而递减;胎肝中Notch1基因mRNA的表达水平有明显的差异,其中E10.5表达量最低,E11.5表达量最高;在各个时相,AGM区与胎肝中Notch1基因mRNA的表达水平差异均有统计学意义(P<0.05)。Notch1蛋白与Notch1基因mRNA的表达规律相同,但除E9.5外,其他3个时相AGM区与胎肝中Notch1蛋白的表达水平差异均无统计学意义(P>0.05)。结论 Notch1信号在造血祖细胞的发育分化中具有重要作用,规律是由AGM区向胎肝区迁移。Objective To investigate the expression of Notch1 signal at early stage of development of heatopoietic system of mouse embryos.Methods The expression levels of Notch1 mRNA and protein in aorta-gonad-mesonephros(AGM) region and liver of mouse embryos at ages of 9.5(E9.5),10.5(E10.5),11.5(E11.5) and 12.5 d(E12.5) were determined by RT-PCR and Western blot respectively.Results The expression level of Notch1 mRNA in AGM region was relatively high and decreased with the increasing gestational period.The expression level of Notch1 mRNA in liver of mouse embryos at various ages showed significant difference,which was the lowest in E10.5 and the highest in E11.5.The expression levels of Nothc1 mRNA in both AGM region and liver at various stages showed significant difference(P 0.05).The regularity of Notch1 protein expression was the same as that of Notch1 mRNA expression.However,the expression levels of Notch1 protein in AGM region and liver of mouse embryos at various stages except E9.5 showed no significant difference(P 0.05).Conclusion Notch1 signal played an important role in development and differentiation of haematopoietic progenitor cells,of which the regularity was migration from AGM region to liver of embryos.
