摘 要:目的分离并鉴定喉癌患者癌组织和癌旁组织的差异表达蛋白质,探讨喉癌发生、发展的分子机制。方法收集喉癌患者癌组织和癌旁组织标本各10例,二维电泳分离总蛋白质,鉴定有明显表达差异的蛋白质点,并对其进行质谱分析。结果获得了分辨率和重复性均良好的凝胶蛋白质图谱,筛选出46个有明显表达差异的蛋白质点,共有17个蛋白质点被成功鉴定,其中在喉癌组织中低表达的有5个,高表达的有12个。结论喉癌患者癌组织蛋白质的表达相对于癌旁组织存在明显差异,差异蛋白质的鉴定有望成为筛选喉癌特异性肿瘤标志物及药物治疗靶点的依据。Objective To explore the molecular mechanism of occurrence and progression of human laryngeal cancer by isolating and identifying the differentially expressed proteins in human laryngeal cancer and adjacent non-carcinoma tissues.Methods Tissue samples were harvested respectively from cancerous and adjacent normal tissue of 10 laryngeal cancer patients.The expressed proteins in tumor and normal tissues were analyzed by two-dimensional gel electrophoresis.The differentially expressed proteins in tumors were also analyzed by mass spectrometry(MS).Results The proteins extracted from individual sample of both types of tissue were separated on two-dimensional gels.A total of 46 differentially expressed proteins in the tissue of laryngeal cancer were screened and 17 proteins were successfully identified by MS.Among them 5 proteins were low expressed and the other 12 proteins were overexpressed.Conclusion Significant discrepancies of protein expression existed between laryngeal cancer and adjacent normal tissues.The differentially expressed proteins may be valuable for finding the specific tumor markers of laryngeal cancer and the targets for treatment.
