摘 要:[目的]研究适宜嗜热厌氧杆菌X514生长和代谢的氮源培养基添加物。[方法]比较X514在添加不同氮源添加物(有机氮源蛋白胨、酵母提取物和无机氮源NH4C l)的基本培养基中的生长和代谢。[结果]酵母提取物和蛋白胨相对于无机氮源都能促进X514的生长,其最大生长速率是0.150 h-(添加5 g/L的酵母提取物培养基),同时X514在高浓度蛋白胨培养基中(5 g/L)的生长相对于低浓度蛋白胨培养基(0.5 g/L)延迟4 h左右;酵母提取物能有效地促进X514产乙醇,与含等量的蛋白胨培养基相比其乙醇转换率(mol产出乙醇/mol消耗的葡萄糖)平均高30%左右。[结论]酵母提取物可以更高效地促进X514的生长和乙醇产出。[Objective] The aim was to find out the appropriate nitrogen medium supplement for Thermoanaerobacter sp.X514.[Method] After comparing the growth and mechanism of X514 in media added with different nitrogen medium supplements(including the organic medium supplements:trptone and yeast extract and inorganic medium supplement NH4Cl),a suitable medium supplement for X514 was found.[Result]The grwoth results in various media added with 5 g/L glucose showed that trptone and yeast extract could promote the growth of X514 compared to NH4Cl;the maximum growth rate was 0.15 h-(the basal medium added with 5 g/L yeast extract);and the growth of X514 in the medium with high concentration of trytone(5 g/L) was delayed for 4 hours compared to it in the medium with lower concentration of tryptone(0.5 g/L).Finally,after analyzing the product distribution in those media,yeast extract was more effective in promting the ethanol production and the ethanol conversation rates(mol ethanol produced/ mol glucose consumed) in media added with yeast extract was 30% more than those in media added with equal amount of tryptone on average.[Conclusion]Through those experiments,it was found that compared with the equal amount of tryptone,yeast extract was more effective in promoting the growth and ethanol production of X514.
